HPLC测定人参-天麻药对不同配比中的人参皂苷Re

Determination of ginsenoside Re in Panax ginseng-Gastrodia elata drugs in different proportions by HPLC

目的采用HPLC法测定不同配比的人参-天麻药对水煎液中人参皂苷Re的含量,探讨人参-天麻药对配伍比例的合理性。方法设置不同剂量配比的人参-天麻药对(1∶0、1∶1、2∶1、3∶1、4∶1、1∶2、1∶3、1∶4),采用水煎法提取;采用Inert Sustain C18色谱柱,流动相为乙腈-0.05%磷酸(20∶80),流速1 mL·min^-1,检测波长203 nm,进样量10μL。结果25~500μg·mL^-1人参皂苷Re与峰面积的线性关系良好,回归方程为:Y=299.55X-590.89(r2=0.9994),且重复性、稳定性和回收率均良好,RSD≤2.19%。人参与天麻的比例为1∶0、1∶1、2∶1、3∶1、4∶1、1∶2、1∶3、1∶4的混煎液中人参皂苷Re的含量分别为3.147‰±0.033‰、2.394‰±0.052‰、2.779‰±0.040‰、2.759‰±2.156‰、、3.006‰±0.028‰、1.857‰±0.028‰、1.566‰±0.035‰、1.299‰±0.061‰。结论人参-天麻药对配伍中人参皂苷Re的含量随着天麻比重的增加而逐渐降低,且天麻的比重越高,人参皂苷Re的含量下降越明显。说明天麻配伍人参可以降低人参中人参皂苷Re的含量。

OBJECTIVE To detect ginsenoside Re in the water decoction of Panax ginseng-Gastrodia elata by HPLC and to discuss the rationality of the ratio of Panax ginseng-Gastrodia elata.METHODS Ginseng-Gastrodia pairs(1∶0,1∶1,2∶1,3∶1,4∶1,1∶2,1∶3,1∶4)were extracted by water decoction.Chromatographic conditions performanced with an InertSustain C18 column,and the acetonitrile-0.05%phosphoric acid(20∶80)was adopted as mobile phase with flow rate of 1 mL·min^-1.The detection wavelength was 203 nm with the injection volume of 10μL.RESULTS The linear regression equation was Y=299.55X-590.89(r2=0.9994).And the reproducibility,stability and recovery were all good,with the RSD less than 2.19%.The contents of ginsenoside Re in the mixed decoction in which the proportion of Panax ginseng to Gastrodia elata of 1∶0,1∶1,2∶1,3∶1,4∶1,1∶2,1∶3,1∶4 were 3.147‰±0.033‰,2.394‰±0.052‰,2.779‰±0.040‰,2.759‰±2.156‰,3.006‰±0.028‰,1.857‰±0.028‰,1.566‰±0.035‰and 1.299‰±0.061‰,respectively.CONCLUSION The content of ginsenoside Re in the compatibility of Panax ginseng and Gastrodia elata decrease gradually with the increase of the proportion of Gastrodia elata.The higher proportion of Gastrodia elata is the more obvious decrease of the content of ginsenoside Re will be,and the difference appears statistically significant.It shows that Gastrodia elata combined with Panax ginseng can reduce the content of ginsenoside Re.
