补肾填精益髓法对脑出血大鼠模型NSE和GFAP影响

The Effects of Kidney-Tonifying and Lean Pulp-Filling Method on GFAP and NSE in Rats with Cerebral Hemorrhage

摘要 目的:研究补肾填精益髓法对脑出血大鼠胶质纤维酸性蛋白(GFAP)和神经元特异性烯醇化酶(NSE)的影响。方法:125只SD大鼠随机分为5组,分别为假手术组、模型组、中药组、西药组、中西药联合组,各组25只,采用立体定向仪自体血注入法制作大鼠脑出血模型,各治疗组分别灌服相应中药浓缩液及吡拉西坦片浓缩液,各组于造模成功后24 h、治疗第7、14、21、28天各处死5只大鼠,采用免疫法测大鼠脑组织尼氏小体变化及GFAP表达,酶联免疫法检测大鼠血清NSE含量。结果:造模24 h后模型组尼氏小体数量减少、脑组织GFAP表达较假手术组显著增高(P<0.01),表明造模成功。治疗开始后与模型组相比,西药组、中西药联合组大鼠血清NSE浓度及脑组织GFAP表达在治疗第14天开始显著下降(P<0.01);中药组大鼠血清NSE在治疗第14天开始显著下降(P<0.01),脑组织GFAP表达在第28天显著下降(P<0.01)。与中药组相比,西药组、中西药联合组大鼠血清NSE浓度及脑组织GFAP表达均在治疗第14天开始显著下降(P<0.05)。与西药组相比,中西药联合组在治疗第28天大鼠血清NSE浓度及脑组织GFAP表达显著下降(P<0.05)。结论:补肾填精益髓法可显著降低脑出血大鼠模型血清NSE浓度及脑组织GFAP蛋白表达,中西药联合治疗效果优于单独中药或西药治疗。 Objective:To study the effects of Kidney-tonifying and Lean Pulp-filling Method on Glial Fibrillary Acidic Protein(GFAP)and Neuron Specific Enolase(NSE)in rats with cerebral hemorrhage. Methods:125 SD rats were randomly divided into 5 groups:sham operation group,model group,Chinese medicine group,western medicine group,Chinese and Western medicine combined group,25 in each group. Rat brain was made by autologous blood injection method Bleeding model,the treatment group was given the Chinese medicine concentrate or piracetam concentrate respectively. Rat in each group was killed in 1 th,7 d,14 d,21 d and 28 d. Immunoassay was used to measure the change of nissl bodies and GFAP expression in brain tissue of rats,and the serum NSE content in rats was detected by enzyme-linked immunosorbent assay. Results:After 24 hours of modeling successfully,the serum NSE concentration and GFAP expression in the model group were significantly higher than those in the sham operation group(P<0.01),indicating successful modeling. Compared with the model group,the serum NSE concentration and GFAP expression in the western medicine group and the Chinese and Western medicine combined group began to decrease significantly on the 14 th day after treatment(P<0.01). The serum NSE of the Chinese medicine group began to decrease significantly on the 14 th day of treatment(P<0.01),GFAP expression in brain tissue decreased significantly on day 28(P<0.01). Compared with the traditional Chinese medicine group,the serum NSE concentration and GFAP expression in the western medicine group and the Chinese and Western medicine combined group began to decrease significantly on the 14 th day of treatment(P<0.05).Compared with the western medicine group,the serum NSE concentration and GFAP expression in the brain tissue of the Chinese and Western medicine combined group decreased significantly on the 28 th day of treatment(P<0.05). Conclusion:Kidney-tonifying and Lean Pulp-filling Method can significantly reduce serum NSE concentration and brain tissue GFAP protein expression in rats with cerebral hemorrhage. The combination of Chinese and Western medicine is better than traditional Chinese medicine or western medicine.
