辛通畅络法对局灶节段性肾小球硬化大鼠内皮素-1及一氧化氮表达的影响 被引量:4

Effectof Xintong Changluo Method on Endothelin-1 and Nitric Oxide in Rats with Focal Segmental Glomerulosclerosis

摘要 目的:观察辛通畅络法中药复方肾络宁对局灶节段性肾小球硬化(FSGS)型阿霉素肾病大鼠模型内皮素-1、一氧化氮、肾小球毛细血管袢的影响。方法:采用左肾切除加阿霉素注射法诱发SD大鼠FSGS模型,依体重随机分模型组、西药组、中药组,另设空白对照组。西药组灌服贝那普利混悬液,中药组灌服肾络宁,模型组及对照组灌服等量生理盐水,于治疗第12周末,检测血浆ET-1、血清NO,肾小球毛细血管袢横截面积和毛细血管腔开放程度、肾小球系膜基质面积。结果:西药组、中药组血浆ET-1较模型组降低(P<0.05),西药组、中药组血清NO较模型组升高(P<0.05)。中药组、西药组肾小球毛细血管袢横截面积、肾小球毛细血管腔开放程度,均较模型组增加(P<0.01、P<0.05)。中药组、西药组肾小球系膜基质面积及相对面积较模型组减轻(P<0.01)。结论:辛通畅络法复方肾络宁对局灶节段性肾小球硬化型阿霉素肾病大鼠具有降低血浆ET-1表达,增加血清NO表达,减轻肾小球系膜基质增生,改善肾小球毛细血管袢损害。 Objective:To observe the effect of Shenluoning from Xintong Changluo therapeutic principle on expressions of endothelin-1(ET-1)and nitric oxide(NO),glomerular capillary loop andglomerular mesangial matrix in focal segmental glomerulosclerosis(FSGS)model rats.Methods:FSGS model was induced in SD rats by injecting Adriamycin and left nephrectomy.They were randomly divided into the model group,the Western medicine groupand the Chinese medicine group according to body weight,12 rats in each group.Besides,another 12 rats was recruited as the control group.By gastrogavage,Benazepril(0.33 mg/100 g)was given to rats in the Western medicine group,while Shenluoning(3.5 g/100 g)was given to rats in the Chinese medicine group and normal saline was given to rats in the control group and the model group.After 12 weeks,we detected ET-1 and NO in serum,measured glomerular capillary loop area(μm^2),glomerular capillary lumen diameter(μm),glomerular mesangial matrix area by patho-image analysis system.Results:Compared with the model group at the same time,ET-1 of Western medicine group and Chinese medicine group decreased(P<0.05).NO of Western medicine group and Chinese medicine group increased(P<0.05).Glomerular capillary loop area and glomerular capillary lumen diameter improved in Western medicine group and Chinese medicine group(P<0.05,P<0.01).The glomerular mesangial matrix area and glomerular mesangial matrix relative area decreased in Western medicine group and Chinese medicine group(P<0.01).Conclusion:Xintong Changluo method Shenluoning could reduce glomerular capillary loop damage and decrease glomerular mesangial matrix hyperplasia in FSGS rats,which may be related with the depression of ET-1 and increase of NO.
