补中益气汤对化疗性大鼠消化道不良反应的防治效果及机制探讨

Prevention and treatment effect of Buzhong Yiqi Decoction on chemotherapy-induced gastrointestinal adverse reactions in rats and its mechanism

目的:观察补中益气汤对化疗性大鼠消化道不良反应的防治作用并探讨其作用机制。方法:56只Wistar大鼠分为空白对照组、补中益气汤正常对照组、顺铂模型组、昂丹司琼阳性对照组、补中益气汤低剂量组、中剂量组和高剂量组7组。后5组腹腔注射顺铂6 mg/kg进行造模,各组分别灌胃生理盐水、补中益气汤1.6 g/kg、生理盐水、顺铂模型组、昂丹司琼1.3 mg/kg、补中益气汤1.6 g/kg、补中益气汤3.2 g/kg、补中益气汤6.4 g/kg,注射容积均为10 ml/kg。记录大鼠高岭土消耗量后,取其血清检测胃肠激素水平,观察病理组织形态学变化。结果:补中益气汤和昂丹司琼对于防治大鼠化疗性消化道不良反应均具有良好作用。补中益气汤低、中、高剂量组、昂丹司琼阳性对照组大鼠血清中胃泌素、胃动素水平明显高于顺铂模型组,生长抑素、血管活性肠肽水平明显低于顺铂模型组,且各组间比较差异无统计学意义。结论:补中益气汤对化疗性大鼠消化道不良反应有明显防治作用,其作用机制与改善化疗引起的胃肠激素分泌紊乱有关。

Objective:To observe the therapeutic effect of BuZhong YiQi Decoction for chemotherapy gastrointestinal side effects of rats,and explore its mechanism.Methods:56 Wistar rats were divided into 7 groups.They were blank control group,Buzhong Yiqi Decoction normal control group,cisplatin model group,ondansetron positive control group,Buzhong Yiqi Decoction low dose group,middle dose group and high dose group.Five groups were intraperitoneally injected with cisplatin 6 mg/kg for injection molding.Each group was intragastrically administered,saline,Buzhong Yiqi Decoction 1.6 g/kg,normal saline,cisplatin model group,ondansetron 1.3 mg/kg,Buzhong Yiqi Decoction 1.6 g/kg,Buzhong Yiqi Decoction 3.2 g/kg,Buzhong Yiqi Decoction 6.4 g/kg.The injection volume was 10 ml/kg.In the experiment,the kaolin consumption of the rats was recorded.At the end of the experiment,blood was taken from the inferior vena cava and centrifuged to obtain serum gastrin and detect gastrointestinal hormone levels.The histopathological changes were observed by HE staining.Results:Comparing the pathological sections of gastrointestinal mucosa in each group,it was found that Buzhong Yiqi Decoction and Ondansetron had good effects on the prevention and treatment of adverse reactions of chemotherapy gastrointestinal tract in rats.The serum levels of gastrin and motilin in Buzhong Yiqi Decoction low-dose group,Buzhong Yiqi Decoction medium-dose group,Buzhong Yiqi Decoction high-dose group and Ondansetron positive control group were significantly higher than cisplatin model group.The levels of somatostatin and vasoactive intestinal peptide were significantly lower than those in the cisplatin model group,and there was no statistical difference between the groups.Conclusion:Buzhong Yiqi Decoction has obvious preventive and therapeutic effects on the adverse reactions of gastrointestinal tract in chemotherapeutic rats.Its mechanism of action is related to the improvement of gastrointestinal hormone secretion disorder caused by chemotherapy.
