吉林省动物源食物链中肠球菌污染及检测结果分析 被引量:1

Analysis of contamination and detection results of resistant enterococcus in animal food chain in Jilin province

摘要 目的了解吉林省某猪场中生猪养殖链条中肠球菌污染状况,为进一步耐药性基因的监测提供菌株。方法采集不同日龄的新鲜猪粪(ZF)、猪鼻腔拭子(ZB)、猪舍地板涂抹物(D)、猪舍墙壁涂抹物(Q)、污水(W)、工作人员鼻腔拭子(RB)、周围土壤(T)标本共计138份。采集的样品经处理后进行选择性增菌、分离、生化及PCR鉴定。结果育肥前共采集68份样品,检出肠球菌61株;育肥后70份样品,检出肠球菌51株,共检出112株肠球菌,总阳性率为81.2%。其中阳性率由高到低依次为ZF:95.0%,D:94.4%,Q:83.3%,ZB:77.5%,W:75.0%,T:37.5%,RB:33.3。样品的总检出和猪鼻腔拭子育肥前肠球菌的阳性率明显高于育肥后的样品(P<0.05),表明育肥后猪龄大了其本身对于肠球菌感染的抵抗力增强。通过调查该猪场饲料中既添加了恩拉霉素、蝰乙醇、硫酸粘杆菌素3种抗生素,又添加硫酸铜、硫酸亚铁、氧化锌3种重金属。结论通过监测提示动物源性样品中肠球菌的污染是严重的。通过调查表明我国猪的养殖场饲料中添加抗生素和重金属是普遍的,值得有关部门高度重视。通过本次监测为进一步研究动物源食物链中肠球菌耐药性提供了大量菌株。 Objective To investigate the contamination status of enterococci in pig breeding chain in a pig farm in Jilin province and to provide strains for further surveillance of drug resistance genes.Methods Fresh pig manure(ZF)at different ages;Pig nasal swab(ZB);Pigsty floor coating(D);Pigsty wall daub(Q);Sewage(W);Staff nasal swab(RB)were collected.There were 138 samples of surrounding soil(T).The samples were treated for selective enrichment,isolation,biochemistry and PCR identification.Results Before fattening,68 samples were collected and 61 strains of enterococcus were detected.Fifty-one enterococci strains were detected in 70 samples after fattening.A total of 112 enterococci strains were detected,the total positive rate was 81.2%.The positive rate was ZF:95.0%D:94.4%;Q:83.3%;ZB:77.5%;W:75.0%;T:37.5%;RB:33.3.from high to low.The total detection of samples and nasal swabs fattening enterococci positive rate was significantly higher than the fattening samples(P<0.05),which meant that the older pigs were after fattening,the more resistant they were to enterococcal infection.According to the investigation,three antibiotics,such as enrapycin,viper ethanol and myxobacterium sulfate,as well as three heavy metals,such as copper sulfate,ferrous sulfate and zinc oxide,were added into the feed of the pig farm.Conclusion Our monitoring indicates that the contamination of enterococci in animal-derived samples is serious.According to the investigation,antibiotics and heavy metals are commonly added into the feed of pig farms in China,which deserves high attention of relevant departments.Through this monitoring,a large number of strains of enterococci in the food chain of animal origin have been provided for further research on drug resistance.drug resistance.
