卤醇脱卤酶催化合成(S)-2-溴-1-苯基乙醇的工艺优化

Optimization in Enzymatic Synthesis Process of (S)-2-Bromo-1-phenylethanol by Halohydrin Dehalogenase

摘要 以新型卤醇脱卤酶AbHHDH为催化剂,催化拆分外消旋2-溴-1-苯基乙醇生成光学纯的(S)-2-溴-1-苯基乙醇,通过单因素实验探究了缓冲液pH值、缓冲液浓度、催化剂用量、底物浓度以及温度对催化合成反应的影响。结果表明,在NaH2PO4-Na2HPO4缓冲液pH值为8.0、NaH2PO4-Na2HPO4缓冲液浓度为200 mmol·L^-1、催化剂用量为37.5 g·L^-1、底物浓度为20 mmol·L^-1、温度为28℃的最优工艺条件下反应15 min,(S)-2-溴-1-苯基乙醇的ee值为100%,收率为35.11%。 Using halohydrin dehalogenase from an alphaproteobacterium(AbHHDH)as a catalyst,we synthesized(S)-2-bromo-1-phenylethanol by enantioselective resolution of racemic 2-bromo-1-phenylethanol.Moreover,we investigated the effects of buffer pH value,buffer concentration,catalyst amount,substrate concentration,and temperature on catalytic synthesis reaction by single-factor experiments.The results show that the optimal conditions are determined as follows:the buffer pH value of 8.0,the buffer concentration of 200 mmol·L^-1,the catalyst amount of 37.5 g·L^-1,the substrate concentration of 20 mmol·L^-1,and the temperature of 28℃.Under above conditions for 15 min,the ee value and the yield of(S)-2-bromo-1-phenylethanol reach 100%and 35.11%,respectively.
