骨髓间充质干细胞对溃疡性结肠炎小鼠受损肠黏膜修复作用的探究

Effect of bone marrow mesenchymal stem cells on the repair of damaged intestinal mucosa in mice with ulcerative colitis

目的探讨骨髓间充质干细胞(BMSCs)对溃疡性结肠炎(UC)小鼠模型中肠黏膜的修复作用。方法BALB/F雌性小鼠随机分为正常对照组、实验对照组(DSS+PBS组)、实验组(DSS+BMSCs组),正常对照组正常饮水,其余2组每天饮用4%的葡聚糖硫酸钠(DSS)水溶液,造模第8天所有对照组及实验组分别用磷酸盐缓冲液(PBS)、BMSCs进行干预治疗,并于BMSCs注射后第2、5、9、12、16天处死各组5只小鼠,留取标本。根据疾病活动度(DAI)评分评估疾病的活动情况;免疫组化、ELISA分别检测结肠组织中间质表皮转化因子(c-Met)、表皮生长因子受体(EGFR)、增殖细胞核抗原(PCNA)及肝细胞生长因子(HGF)、肝细胞生长因子激活因子(HGFA)的表达。结果实验组及实验对照组小鼠DAI评分均高于正常对照组,且实验组评分低于实验对照组;干细胞注射后第5天及以后,实验对照组HGF的表达量均明显低于正常对照组;除第2、9天外,实验组HGF的表达与正常对照组差异不大或高于正常对照组,但均明显高于实验对照组;HGFA在实验对照组中表达先下降后上升,但其在3组间的表达无明显规律性差异。实验对照组小鼠结肠组织中c-Met、EGFR、PCNA表达明显低于正常对照组和实验组,实验组的表达略低于正常对照组或相差不大。结论BMSCs移植治疗可能通过促进DSS诱导的UC小鼠受损肠黏膜的细胞增殖来促进黏膜修复。

Objective To iveseigate the repair efet of bone marrow mesenchymal stem cll(BMSCs)on injured intestinal mucosa in a mouse model of ulcerative coitis.Methods BALB/C female mice were randomly divided into three groups.The normal contol goup was regulaly fed and was not fed with dextan sodium sulfate(DSS),while the model group and the experimental group were fed with 4%DSS.After drinking water for 8 days,the ltter two groups were injected with PBS and BMSCs respectively,then 5 mice in each group were sacificed on day2.5、9、12 and 16 after injection.Colon tissues were harvested.Results The Disease Activity Index(DAI)scores of both experimental groups(DSS+BMSCs)and experimental control groups(DSS+PBS)were higher than the nor-mal control goup,and the scores of experimental group were lower than experimental control group.The expression level of HCF in the experimental contol goup was sigifcanly lower than normal contol group on day 5 and be-yond after injection.Except for the day 9,the expression of HCF in the experimental group was not sigifcanly dfferent from that in the normal control group but higher than the experimental contrl group.There was no obvious reglar changes in the epresion of HGFA bewcen the tree goups.The epresss of c-Met,ECFR and PCNAin the colon lsus of the control group was sgifi amly lower than that in the nomal control goup and the exper-mental goup.However,the epresion of the eperinenal group was sighly lower than that in the normal control goup o"r the dference was no obrious.Concusion The study sugestes that BMSCs rsplantation promotes cell proliferation lo repair the damaged colon issues of mice models with uc.
