摘要 目的检测长链非编码RNA(LncRNA)核内小RNA宿主基因20(SNHG20)和微小核糖核酸-495(miR-495)在宫颈癌患者癌组织及癌旁正常组织中的表达水平,分析二者表达相关性及与宫颈癌疾病的关系。方法选取2011年6月至2013年8月在本院诊治并接受手术的宫颈癌患者的癌组织及配对癌旁组织分别设为研究组与对照组,每组74例,采用实时荧光定量PCR技术检测Lnc SNHG20和miR-495表达水平,分析Lnc SNHG20与miR-495表达相关性及与宫颈癌疾病的关系。结果研究组中Lnc SNHG20相对表达量显著高于对照组(P<0.05),miR-495相对表达量显著低于对照组(P<0.05)。Lnc SNHG20和miR-495表达水平与患者FIGO分期、组织分化、肿瘤直径和淋巴结是否转移有关(P<0.05)。Lnc SNHG20高表达组生存率显著低于低表达组(P<0.05),miR-495低表达组生存率显著低于高表达组(P<0.05)。Lnc SNHG20表达与miR-495号负相关(r=-0.839,P<0.01),是影响宫颈癌不良预后发生的独立危险因素(P<0.05)。结论Lnc SNHG20在宫颈癌患者癌组织中高表达,miR-495低表达,二者表达水平负相关,与FIGO分期、组织分化、肿瘤直径和淋巴结是否转移有关,是影响宫颈癌不良预后发生的独立危险因素。 Objective To investigate To detect the expressions of long non-coding RNA(LncRNA)small nucleolar RNA host gene 20(SNHG20)and micro RNA-495(miR-495)in cervical cancer tissues and adjacent normal tissues,and to analyze their correlation with cervical cancer.Methods A total of 74 patients with cervical cancer who were treated in our hospital from June 2011 to August 2013 were enrolled in the study,whose cancer tissues(research group)and adjacent normal tissues(control group)were collected.The expression levels of Lnc SNHG20 and miR-495 were detected by real-time quantitative PCR,and the correlation between the expressions of Lnc SNHG20 and miR-495 as well as their relationship with cervical cancer was analyzed.Results The relative expression levels of Lnc SNHG20 in research group were significantly higher than those in control group(P<0.05),however,the relative expression levels of miR-495 were significantly lower than those in control group(P<0.05).Lnc SNHG20 and miR-495 expression levels were correlated with FIGO stage,tissue differentiation,tumor diameter and lymph node metastasis(P<0.05).The survival rate in Lnc SNHG20 high expression group was significantly lower than that in low expression group(P<0.05),and the survival rate in miR-495 low expression group was significantly lower than that in high expression group(P<0.05).The expression of Lnc SNHG20 was negatively correlated with that of miR-495(P<0.01),which was an independent risk factor for poor prognosis of patients with cervical cancer(P<0.05).Conclusion Lnc SNHG20 is highly expressed in cancer tissues of patients with cervical cancer,however,miR-495 is lowly expressed,there is a negative correlation between their expression,moreover,which are related to FIGO stage,histological differentiation,tumor diameter and lymph node metastasis,which are independent risk factors affecting the incidence od adverse prognosis of patients with cervical cancer.
