摘要
目的检测非受体型蛋白酪氨酸磷酸酶12(protein tyrosine phosphatase,non-receptor type 12,PTPN12)及PTPN12基因mRNA在肾细胞癌(renal cell carcinoma,RCC)组织及相应正常组织中的表达。方法运用免疫组化P-V染色法检测RCC组织中及相应正常组织中PTPN12蛋白的表达,采用实时定量基因扩增荧光检测系统(real-time quantitative polymerase chain reaction detecting system,RT-qPCR)检测RCC组织及相应正常组织中PTPN12基因mRNA相对表达量,并分析其与各临床病理资料之间的关系。结果在RCC组织中PTPN12蛋白表达的阳性率及mRNA的相对表达量显著低于相应正常组织,差异有统计学意义(P<0.05)。RCC组织中PTPN12蛋白及mRNA表达情况与患者的发病年龄、性别、肿瘤直径及临床分期之间均无明显关联(P>0.05)。结论RCC中PTPN12的低表达可能与RCC的发生有关,但可能与患者发病年龄、性别、肿瘤直径及临床分期无关。
Objective To detect the expression of protein tyrosine phosphatase,non-receptor type 12(PTPN12)and PTPN12 mRNA in renal cell carcinoma(RCC)tissues and corresponding normal tissues.Methods The immunohistochemical P-V staining was used to detect the expression of PTPN12 protein in renal cell carcinoma(RCC)tissues and corresponding normal tissues.The real-time quantitative polymerase chain reaction detecting system(RT-qPCR)was used to detect the relative expression levels of PTPN12 mRNA in RCC tissues and corresponding normal tissues.And the correlation between the expression of FAM83h in renal cell carcinoma and clinicopathological data was nalyzed.Results The positive rate of PTPN12 protein expression and the relative expression levels of PTPN12 mRNA in RCC tissues were significantly lower than those in normal tissues(P<0.05).There was no significant correlation between the expression levels of PTPN12 protein and mRNA in RCC tissues and clinicopathological data including patient’s age and gender,tumor diameter and clinical stage(P>0.05).Conclusion The low expression of PTPN12 in RCC may be related to the occurrence of RCC,however,which may not be related to patient’s age and gender,tumor diameter and clinical stage of the patients.
