摘要 目的:研究ActivinA对人胚胎干细胞体外神经外胚层诱导效率的影响。方法:对照组细胞在神经外胚层诱导培养基中培养5天;实验组细胞在神经外胚层诱导培养基中培养2天后换成含100 ng/mL Activin A培养基中继续诱导3天。通过免疫荧光检测细胞中PAX6蛋白的表达情况,利用RT-qPCR检测PAX6、POU5F1等基因的表达水平。结果:实验组中PAX6阳性的细胞明显多于对照组细胞,其PAX6基因表达结果也证实了这一点,POU5F1基因表达下调更为显著。结论:Activin A能够显著促进胚胎干细胞向神经外胚层细胞的转变。 Objective: To investigate the effects of Activin A on neuroectoderm differentiation from human embryonic stem cells(hESCs). Methods: Cells in the control group were cultured in neural induction medium for 5 days. Cells in the experimental group were cultured in the same induction medium only for 2 days and then replaced with the medium containing 100 ng/mL Activin A for 3 days. The protein level of PAX6 was detected by immunofluorescence, and the mRNA abundance of PAX6 and POU5 F1 was detected by RT-qPCR. Results: We detected more PAX6-positive cells in the experimental group in contrast to the control group, which was confirmed by RT-qPCR. Expression of POU5 F1 was significantly downregulated with the addition of Activin A. Conclusions: Activin A can significantly promote the generation of neuroectodermal cells.
