摘要
目的分析靶向抑制头颈部鳞癌细胞中TANK结合酶1(TANK binding,kinase 1,TBK1)表达后细胞增殖与凋亡水平改变。方法采用转染TBK1 siRNA靶向抑制人下咽鳞癌细胞FaDu、人舌鳞癌细胞Cal-27中TBK1的表达,用CCK-8、流式细胞术、Western Blot等方法检测FaDu及Cal-27中靶向抑制TBK1表达前后细胞的增殖及凋亡水平;克隆形成实验及细胞增殖实验(CCK-8法)检测梯度浓度氨来呫诺处理后FaDu及Cal-27细胞增殖改变。结果Western Blot结果显示,转染后FaDu及Cal-27细胞中TBK1蛋白表达显著抑制;细胞增殖实验结果显示,靶向抑制TBK1蛋白表达后,FaDu及Cal-27的增殖显著抑制;流式细胞术检测显示FaDu及Cal-27细胞凋亡显著增加;克隆形成及细胞增殖实验结果表明FaDu及Cal-27细胞的增殖随着氨来呫诺浓度增加呈现显著下降(P<0.05)。结论靶向抑制TBK1蛋白表达后,舌及下咽鳞癌细胞的增殖能力明显降低,细胞凋亡显著增加;TBK1抑制剂氨来呫诺对于高表达TBK1的舌及下咽鳞状细胞癌的增殖具有抑制作用。
Objective To study the effects of targeted inhibit of TANK binding kinase 1 expression on cell proliferations and apoptosis in tongue and pharynx squamous carcinoma.Methods TBK1 siRNA was transfected to inhibit the expression of TBK1 in human tongue and pharynx squamous cell carcinoma cells(FaDu,Cal-27).CCK-8,flow cytometry and Western Blot were used to detect the proliferation and apoptosis of tongue and pharynx squamous cell carcinoma cells before and after transfection.The proliferation of FaDu and Cal-27 cells treated with gradient concentration of amlexanol was detected by colony formation assay and CCK-8 assay.Results Western Blot results showed that the expression of TBK1 protein in FaDu and Cal-27 cells was significantly inhibited after transfection;CCK-8 results showed that the proliferation of FaDu and Cal-27 cells was significantly inhibited;flow cytometry results showed that the apoptosis of FaDu and Cal-27 cells increased significantly;cloning formation experiment and CCK-8 experimental results showed that the apoptosis of FaDu and Cal-27 cells increased significantly.The results showed that the proliferation of FaDu and Cal-27 decreased significantly with the increase of ammonia concentration(P<0.05).Conclusion After targeting inhibition of TBK1 protein expression,the proliferation ability and apoptosis of FaDu and Cal-27 tongue and pharynx squamous cell carcinoma cells decreased significantly.Amlexanol can inhibit the proliferation of tongue and pharynx squamous cell carcinoma with high expression of TBK1.
