厚朴提取物透皮吸收制剂抗腹泻作用的实验研究

Anti-diarrhea Effect of transdermal absorption preparation of Magnolia Extract

目的研究自制厚朴提取物透皮吸收制剂的抗腹泻作用。方法将ICR小鼠分为厚朴提取物透皮吸收制剂低、中、高(1、2、4 g/kg)剂量组,阳性对照组(洛哌丁胺,1.95 mg/kg)、模型组、空白对照组,每组8只。分别以蓖麻油(每只2 mL)、番泻叶煎剂(生药0.1 g/mL,20 mL/kg)造腹泻小鼠模型,以1~6 h内每小时湿粪计数及累计湿粪计数为检测指标,观察厚朴提取物透皮吸收制剂对腹泻小鼠的作用。结果蓖麻油显著增加小鼠湿粪粒数及累计湿粪粒数(P<0.001),可被洛哌丁胺(1.95 mg/kg)、厚朴提取物透皮吸收制剂高剂量(4 g/kg)、中剂量(2 g/kg)降低,作用起效时间分别为1、2、2 h,维持时间约为4、3、3 h,低剂量(1 g/kg)未见明显抗腹泻作用。番泻叶显著增加小鼠湿粪计数与累计湿粪粒数(P<0.001),可被洛哌丁胺(1.95 mg/kg)、厚朴提取物透皮吸收制剂高剂量(4 g/kg)、中剂量(2 g/kg)降低,作用起效时间分别为1、2、4 h,维持时间约为4、2、1 h,低剂量(1 g/kg)未见明显抗腹泻作用。结论厚朴提取物透皮吸收制剂可显著改善蓖麻油和番泻叶所致腹泻,具有良好的抗腹泻作用。

Objective:To study the anti-diarrhea effect of self-made transdermal absorption preparation of Magnolia Extract(TME). Methods:ICR mice were divided into low,medium,and high(1,2,4 g/kg)dose of TME groups,positive control group(Loperamide,1.95 mg/kg)and blank control group,with 8 mice in each group. Diarrhea model was induce by castor oil(2 mL/kg)or decoction of senna(0.1 g/mL of crude drug,20 mL/kg),using counts of damp excrement per hour and cumulative counts of damp excrement in 1~6 h as indicators,to observe the effect of TME on diarrhea mice. Results:Castor oil(2 mL/kg)significantly increased the counts of damp excrement per hour and cumulative counts of damp excrement(P < 0.001),which was decreased by Loperamide(1.95 mg/kg),MTE in high(4 g/kg)and medium(2 g/kg)dose,with 1 h,2 h,2 h of effective time and 4 h,3 h,3 h of maintenance time respectively,and no anti-diarrhea effect was observed in MTE in low(1 g/kg)dose. Decoction of Senna significantly increased the counts of damp excrement per hour and cumulative counts of damp excrement(P < 0.001),which was decreased by Loperamide(1.95 mg/kg),MTE in high(4 g/kg)and medium(2 g/kg)dose,with 1 h,2 h,4 h of effective time and 4 h,2 h,1 h of maintenance time respectively,and no anti-diarrhea effect was observed in MTE in low(1 g/kg)dose. Conclusion:TME can significantly improve castor oil-and senna-induced diarrhea,suggesting well anti-diarrhea effect.
