摘要
目的:研究益母草挥发油对不同发育阶段斑马鱼胚胎毒性作用。方法:将1.5、3.125、6.25、12.5、25.0、50.0、100.0μg/mL的益母草挥发油处理受精后2 h(2 hpf)的斑马鱼胚胎,在12、24、48、72、96 hpf时间点记录斑马鱼胚胎发育情况。结果:益母草挥发油对斑马鱼胚胎具有致死性(12、24、48、96 hpf的LC 50分别为42.79±2.02、36.97±1.91、31.92±0.79、6.98±0.88μg/mL)和致畸性(48、72、96 hpf的TC 50分别为6.31±0.49、3.56±0.48、1.58±0.19μg/mL),同时使斑马鱼胚胎心率减慢,身体形态发生变化,且毒性随着时间和浓度的增加而增强。结论:益母草挥发油对斑马鱼胚胎发育有一定的毒性,为益母草挥发油临床安全应用提供参考。
Objective:To study the toxicity of essential oil from motherwort(Leonurusjaponicus)on zebrafish embryos at different developmental stages.Methods:Zebrafish embryos of 2 hours post fertilization(hpf)were treated with1.5,3.125,6.25,12.5,25.0,50.0,100.0μg/mL motherwort essential oil,then embryonic development of zebrafish was recorded at 12、24、48、96 hpf.Results:The motherwort essential oil had lethality(LC50 of 12,24,48,96 hpf were 42.79±2.02,36.97±1.91,31.92±0.79,6.98±0.88μg/mL,respectively.)and teratogenicity(TC50 of 48,72,96 hpf were 6.31±0.49,3.56±0.48,1.58±0.19μg/mL,respectively.)to zebrafish embryos.It also slowed down the heart rates of zebrafish embryos,changed the body shape,and increased the toxicity with the increase of time and concentration.Conclusion:The toxicity of motherwort essential oil to zebrafish embryonic development is studied,which can provide reference for clinical safe application of motherwort essential oil.
