热敏灸对慢性萎缩性胃炎模型大鼠胃肠激素的影响 被引量:8

Effects of Heat-sensitive Moxibustion on Gastrointestinal Hormones in Rats with Chronic Atrophic Gastritis

摘要 目的:观察热敏灸对慢性萎缩性胃炎(CAG)模型大鼠血清胃泌素(GAS)、生长抑素(SS)、胃动素(MTL)及白细胞介素(1L-6)的影响,探讨其可能的作用机制。方法:66只雄性SD大鼠,采用随机数字表法随机分为空白组、造模组。空白组不予干预,造模组采用复合病因造模法造模12W,随机分为模型组、维酶素组、艾灸组,艾灸组艾灸“中脘”穴40min,艾灸后按2mL/kg/d灌服0.9%NaCl,根据艾灸过程中大鼠尾温变化分为热敏灸组和非热敏灸组,维酶素组灌服等量维酶素,1次/d,共28d。ELISA法检测血清GAS、SS、MTL、1L-6表达,光镜下观察胃黏膜组织病理学变化。结果:①治疗后,与空白组相比,模型组血清GAS、SS表达均降低(P<0.05,P<0.01);与模型组相比,热敏灸组及维酶素组血清GAS表达均升高(P<0.05,P<0.01),热敏灸组血清SS表达升高(P<0.05),热敏灸组在提高血清GAS、SS表达方面优于非热敏灸组(均P<0.05),但与维酶素组疗效相当。②与空白组相比,模型组血清MTL、1L-6表达均升高(P<0.05,P<0.01);与模型组相比,非热敏灸组、热敏灸组、维酶素组血清MTL、1L-6表达均降低(P<0.05,P<0.01);热敏灸组在降低血清MTL、1L-6表达方面优于非热敏灸组(均P<0.05),但与维酶素组疗效相当。结论:热敏灸可能通过上调CAG大鼠GAS及SS的水平,下调MTL及炎症因子IL-6的水平,从而调节胃肠激素的分泌,改善胃黏膜血流量,抑制炎症因子产生,减轻胃黏膜损伤达到治疗CAG的目的。 Objective:To observe the effects of heat-sensitive moxibustion on GAS,SS,MTL and 1L-6 in rats with chronic atrophic gastritis(CAG),and to explore the possible mechanism for treating CAG.Methods:According to random number table,66SD male rats were randomLy divided into the blank group and the model group.The rats in the blank group did not intervene,the rats in the model group were treated with the composite etiology model method to establish CAG model for 12 weeks,they were randomLy divided into the model group,vitacoenzyme group and moxibustion group,the rats in the moxibustion group were treated with moxibustion at“Zhongwan”acupoint for 40 min,after moxibustion,2mL/kg/d was treated with intragastric administration of 0.9%NaCl,according to the change of tail temperature during moxibustion,the rats were divided into heat-sensitive moxibustion group and non heat-sensitive moxibustion group,the rats in the vitacoenzyme group were treated with identical volume of vitacoenzyme,once a day for 28 days.The expressions of serum GAS,SS,MTL,1L-6 were detected by enzyme linked immune sorbent assay(ELISA),optical microscope was applied to observe the histopathological changes of gastric mucosa.Result:①After treatment,compared with the blank group,the expression of serum GAS and SS in the model group were significantly decreased(P<0.05,P<0.01);Compared with the model group,the expression of serum GAS in the heat-sensitive moxibustion group and the vitacoenzyme group was increased(P<0.05,P<0.01),The serum SS expression of the heat-sensitive moxibustion group was increased(P<0.05),and the heat-sensitive moxibustion group was superior to the non heat-sensitive moxibustion group in terms of increasing the expression of serum GAS and SS(both P<0.05),but the effect was similar to that of the vitacoenzyme group.②Compared with the blank group,the expression of serum MTL and 1L-6 in the model group were significantly increased(P<0.05,P<0.01);The non heat-sensitive moxibustion group,heat-sensitive moxibustion group and vitacoenzyme group can all reduce the expressions of serum MTL,1L-6 compared with the model group(P<0.05,P<0.01).The heat-sensitive moxibustion group was superior to the non heat-sensitive moxibustion group in reducing the expression of serum MTL and 1L-6(both P<0.05),but the effect was similar to that of the vitacoenzyme group.Conclusion:Heat moxibustion maybe up-regulate GAS and SS levels and down-regulate MTL and IL-6 levels in CAG rats,thereby regulating the secretion of gastrointestinal hormones,improving the blood flow of gastric mucosa,inhibiting the production of inflammatory factors,and reducing the damage of gastric mucosa.And this is why heat moxibustion can treat CAG.
