DNA甲基转移酶3(DNMT 3)在乳腺癌患者中的表达及意义

Detection of DNMT 3 genes expressions in breast cancer and then evaluating their clinical relevance

目的 检测DNA甲基转移酶3(DNA methyltransferase 3,DNMT 3)基因在乳腺癌患者外周血中的表达,并分析其临床意义。方法 采用实时定量PCR(RT-PCR)检测60例乳腺癌患者(实验组)及50例体检健康女性(对照组)外周血中DNMT 3A、DNMT 3B、DNMT 3L基因的表达,Western blot检测两组间DNMT 3A、DNMT 3B、DNMT 3L蛋白表达,并分析它们与各临床特征之间的关系。结果 实验组DNMT 3A表达(16.592±2.350)较对照组(4.868±3.014)显著升高,差异有统计学意义(t=4.499,P<0.01);实验组DNMT 3B表达(10.926±3.353)较对照组(3.350±2.2367)显著升高,差异有统计学意义(t=13.642,P<0.01);实验组DNMT 3L表达(8.318±5.707)较对照组(7.373±5.680)比较无统计学差异(t=0.866,P=0.388);在不同临床参数的比较中,仅DNMT 3A的表达在组织学类型上有统计学差异(G1Vs G2,P=0.017;G2Vs G3,P=0.018);实验组DNMT 3A和DNMT 3B蛋白表达较对照组显著增加,而DNMT 3L蛋白表达在两组间无差异。结论 DNMT 3A和DNMT 3B在乳腺癌患者外周血中高表达,而DNMT 3L与乳腺癌患者外周血中表达不显著。

Objective To investigate the expressions of DNA methyltransferase 3(DNMT 3)genes in peripheral blood of breast cancer patients,then evaluating their clinical relevance.Methods Real-time quantitative PCR was used to detect the expressions of DNMT 3 A,DNMT 3 B,DNMT 3 Lgenes in peripheral blood of 60 patients with breast cacer(experimental group)and 50 healthy women(control group),the proteins of DNMT 3 A,DNMT 3 B,DNMT 3 Lwere detected using Western blot between two groups,then the relations between the DNMT 3 A,DNMT 3 B,DNMT 3 Lexpressions and clinical pathological parameters were analyzed.Results There was statistically significant difference between two groups(t=4.499,P<0.01)because of the expressions of DNMT 3 Ain peripheral blood were significantly higher in experimental group(16.592±2.350)than that in control group(4.868±3.014),The expressions of DNMT3 Bwere also statistically significant difference between two groups(t=13.642,P<0.01:10.926±3.353 Vs 3.350±2.2367),but the expressions of DNMT 3 Lwere not statistically significant difference between two groups(t=0.866,P=0.388:8.318±5.707 Vs 7.373±5.680).The expressions of DNMT 3 Aand DNMT 3 Bproteins were higher in experimental group than that in control group,but the DNMT 3 Lproteins were not detected in two groups.DNMT 3 A expressions were just related with the grade of tissues in all pathological characteristics(G1 Vs G2,P=0.017;G2 Vs G3,P=0.018).Conclusion DNMT 3 Aand DNMT 3 Bwere overexpressed in peripheral blood of breast cancer patients,but the expresstions of DNMT 3 Lwere not significant difference in peripheral blood of breast cancer patients.
