摘要
目的:探究青龙衣有效组分对人胃癌BGC-823裸鼠肿瘤生长的抑制作用,探讨其作用机制。方法:采用细胞悬液植入法建立人胃癌BGC-823裸鼠原位移植肿瘤模型。将成模裸鼠随机分为模型组、消癌平药组和青龙衣有效组分高剂量组、低剂量组,各给药组均给予相对应药物灌胃,1次/d,连续15 d。通过免疫组化法测定各分组原位移植瘤瘤体中COX-2的表达情况。通过ELISA法测定各分组血清中PGE-2的表达。结果:与模型对照组比较,青龙衣有效组分低、高剂量组对裸鼠的胃癌原位移植瘤均有不同的抑制作用,与模型组比较,通过HE染色表明,随着药物浓度的降低,凋亡比率增加;其瘤体组织COX-2蛋白表达程度均明显下降(P<0.05);裸鼠血清中的PGE-2的含量均明显下降(P<0.05)。结论:青龙衣有效组分具有抑制胃癌侵袭转移的作用,其作用机制可能与下调COX-2蛋白的表达水平和下降的PGE-2含量有关,其中,高剂量青龙衣有效组分对其抑制作用最明显。
Objective:To investigate the inhibitory effect of the effective components of Qinglongyi on the tumor growth of human gastric cancer BGC-823 nude mice,and to explore its mechanism.Methods:The human gastric cancer BGC-823 nude mice orthotopic transplantation tumor model was established by cell suspension implantation method.The nude mice were randomly divided into a model group,the anti-cancer drug group and the Qinglongyi active ingredient group were high group low group,and each drug-administered group was given the corresponding drug gavage once a day for 15 days.The expression of COX-2 in tumor was detected in immunohistochemisty way.The expression of PGE-2 in the serum of each group was detected by ELISA.Results:Compared with the model control group,the effective components of Qinglongyi and the high group had different inhibitory effects on the orthotopic transplantation of gastric cancer in nude mice.Compared with the model group,the concentration of the drug decreased with the concentration of the drug.Increase in death rate.The expression of COX-2 protein in tumor tissue was significantly decreased(P<0.05).The content of PGE-2 in the serum of nude mice was significantly decreased(P<0.05).Conclusion:The effective components of Qinglongyi have the effect of inhibiting the invasion and metastasis of gastric cancer.The mechanism of action may be related to the down-regulation of COX-2 protein expression and decreased PGE-2 content,and the high-dose Qinglongyi active component has the most obvious inhibitory effect.
