摘要
植物防御素是抗菌肽家族的主要成员,是植物先天免疫的重要组成部分。它们在结构上相似,但序列差异较大,这决定着它们功能的多样性。通过对不同物种植物防御素的表征,可以为研究植物防御素功能多样性提供材料,为筛选出抗性优良的植物防御素基因用于改良作物品种提供帮助。本研究克隆得到了2个紫苏防御素基因,分别命名为PfPDF1、PfPDF2。生物信息学分析发现,PfPDF1、PfPDF2均属于Ⅰ类植物防御素,都具有一个保守的Knot1结构域。实时荧光定量PCR结果显示,PfPDF1、PfPDF2仅在种子中具有一定的表达。紫苏是常用的食用、药用植物,因此紫苏防御素基因在转基因作物中可能具有良好的抗菌活性以及食品安全性。
Plant defensins are the main members of the antibacterial peptide family and are an important part of plant innate immunity.They are structurally similar,but the sequence differences are large,which determines the diversity of their functions.Through the characterization of plant defensins of different species,it can provide materials for studying the functional diversity of plant defensins,and provide help for screening the plant defensin genes with excellent resistance for improving crop varieties.In this study,two Perilla frutescens defensin genes were cloned and named PfPDF1 and PfPDF2.Bioinformatics analysis found that both PfPDF1 and PfPDF2 belong to classⅠplant defensins,all of which have a conserved Knot1 domain.The results of real-time PCR showed that PfPDF1 and PfPDF2 had only a certain expression in the seed.Perilla is a commonly used edible and medicinal plant,so Perilla frutescens defensin may have good antibacterial activity and food safety after genetically modified crop varieties.
