摘要
海岛棉体细胞胚发生体系中,长时间的细胞培养常会引起再生植株发育发生畸变,从而影响后续正常植株尤其是转基因植株的获得,棉花体细胞胚胎发生存在的缺点是基因型依赖、培养周期长、不正常胚产生效率高,严重制约了棉花基因功能的验证和转基因育种工作。本研究通过体细胞胚培育海岛棉,观察其性状,并对体细胞培育植株和野生型海岛棉细胞分别进行了流式细胞仪检测和SSR分子标记鉴定。结果表明,与野生型植株相比,再生植株表现出诸多畸变表型,且基因组倍性和序列组成上发生了改变。本研究结果部分阐明畸形苗的分子机制,有助于体细胞胚体系正常植株的筛选和培育。
In the somatic embryogenesis of Gossypium barbadense,the growth of regenerated plants is often distorted by long-time cell culture,which affects subsequent obtainment of the normal plants,especially transgenic plants.The disadvantages of somatic embryogenesis of cotton are genotype dependence,long-time culture cycle,high efficiency of abnormal embryo production,which seriously restricts the researches and breeding applications of functional genes in cottton.In this study,the cotton seedlings were cultivated through somatic embryogenesis,and the abnormal phenotypes in these seedlings were observed,and the somatic cell culture and wild-type sea-island cotton genes were checked through flow cytometry assay and the SSR technology respectively.The results showed that compared with wide type cotton,the regenerated plants displayed some abnomal phenotypes,and further researches elucidated the genomic ploidy and sequence composition of the regenerated plants were changed to some extent.The results of this study elucidate the molecular mechanism of some malformed seedlings,which is helpful for the screening and breeding of normal plants in somatic embryogenesis system.
