血府逐瘀汤通过PI3K/Akt通路缓解大鼠心肌缺血再灌注损伤的实验研究 被引量:13

Experimental Study of Xuefu Zhuyu Decoction on Relieving Myocardial Ischemia Reperfusion Injury in Rats through PI3K/Akt Pathway

摘要 目的:研究血府逐瘀汤通过PI3K/Akt通路缓解大鼠心肌缺血再灌注(I/R)损伤的作用。方法:雄性SD大鼠随机分为假手术组、缺血再灌注组、血府逐瘀汤10 g/kg、20 g/kg组、血府逐瘀汤20 g/kg+LY2940020.3 mg/kg组,各组分别灌胃给予相应药物或蒸馏水,血府逐瘀汤20 g/kg+LY2940020.3 mg/kg组灌胃及腹腔注射,连续干预14天后采用左冠状动脉前降支结扎的方式建立心肌I/R模型,比较心肌梗死面积、血清心肌酶含量、心肌组织细胞凋亡率及凋亡基因、信号通路基因的蛋白表达量。结果:缺血再灌注组大鼠的心肌梗死面积、血清LDH、CK、CK-MB的含量、心肌组织细胞凋亡率及Bax、Caspase-3的表达量均明显高于假手术组,心肌组织Bcl-2、p-PI3K、p-Akt的表达量均明显低于假手术组;血府逐瘀汤10 g/kg、20 g/kg组大鼠的心肌梗死面积、血清LDH、CK、CK-MB的含量、心肌组织细胞凋亡率及Bax、Caspase-3的蛋白表达量均明显低于缺血再灌注组,心肌组织Bcl-2、p-PI3K、p-Akt的蛋白表达量均明显高于缺血再灌注组;血府逐瘀汤20 g/kg+LY2940020.3 mg/kg组的心肌梗死面积、血清LDH、CK、CK-MB的含量、心肌组织细胞凋亡率及Bax、Caspase-3的蛋白表达量均明显高于血府逐瘀汤20 g/kg组,心肌组织Bcl-2、p-PI3K、p-Akt的蛋白表达量均明显低于血府逐瘀汤20 g/kg组。结论:血府逐瘀汤具有缓解大鼠心肌缺血再灌注损伤的作用且该作用可能由PI3K/Akt通路的激活所介导。 To study the relieving effect of XuefuZhuyu Decoction on myocardial ischemia-reperfusion(I/R)injury in rats through PI3 K/Akt pathway.Methods:Male SD rats were randomly divided into SHAM group,I/R group,XuefuZhuyu Decoction groups(10 g/kg,20 g/kg)and 20 g/kg XuefuZhuyu Decoction+0.3 mg/kg LY294002 group.Rats were administered with corresponding drugs or distilled water.In 20 g/kg XuefuZhuyu Decoction+0.3 mg/kg LY294002 group,20 mg/kg XuefuZhuyu Decoction was given intragastrically and 0.3 mg/kg LY294002 was injected intraperitoneally.After 14 days of continuous intervention,myocardial I/R model was established by ligation of anterior descending branch of left coronary artery.Myocardial infarction area,the content of serum myocardial enzyme,the apoptotic rate of myocardial cells,protein expressions of apoptosis gene and signaling pathway gene in myocardial tissues were compared.Results:In I/R model group,the myocardial infarction area,serum contents of LDH,CK and CK-MB,the apoptotic rate and the expressions of Bax,Cyt-C and Caspase-3 in myocardial tissues were significantly higher than those in SHAM group.In XuefuZhuyu Decoction groups(10 g/kg,20 g/kg),the myocardial infarction area,serum contents of LDH,CK and CK-MB,the apoptotic rate and the expressions of Bax,Cyt-C and Caspase-3 in myocardial tissues were lower than those in I/R group,the expressions of Bcl-2,p-PI3 K and p-Akt in myocardial tissues were significantly higher than those in I/R group.In 20 g/kg XuefuZhuyu Decoction+0.3 mg/kg LY294002 group,the myocardial infarction area,serum contents of LDH,CK and CK-MB,the apoptotic rate and the expressions of Bax,Cyt-C and Caspase-3 in myocardial tissues were significantly higher than those in 20 g/kg XuefuZhuyu Decoction group,the expressions of Bcl-2,p-PI3 K and p-Akt in myocardial tissues were significantly lower than those in 20 g/kg XuefuZhuyu Decoction group.Conclusion:XuefuZhuyu Decoction can alleviate myocardial ischemia-reperfusion injury in rats,which may be mediated by the activation of PI3 K/Akt pathway.
