环介导等温扩增技术检测弓形虫方法的建立及其临床应用 被引量:7

摘要 目的探究建立环介导等温扩增方法(LAMP)进行弓形虫病的检测及其临床应用。方法根据弓形虫WH6株表面抗原1的基因序列合成特异性LAMP引物,进行预实验并对扩增的相关参数进行优化,建立LAMP检测体系,并对临床上100例IgG或(和)IgM抗体阳性血液标本以及50例阴性参考血液标本进行细胞的DNA提取后分别进行LAMP和PCR检测,比较其结果,探究其敏感度和特异性。结果当基因稀释至10^-8时为LAMP可检测出的最低浓度,当基因稀释至10^-6时为PCR可检测出的最低浓度,LAMP敏感性为PCR的100倍。两者特异性均较高,100例IgG或(和)IgM抗体阳性血液标本中,LAMP检测方式显示100例(100%)出现阳性反应,而PCR方法检测中74例(74%)出现阳性反应,两种检测方式结果相比具有统计学差异(P<0.05)。结论 LAMP法检测弓形虫感染具有快速、便捷、灵敏度高、特异性强等优点,具有广阔前景,值得临床上推广。
