缺氧诱导因子1α和自噬相关蛋白Beclin-1在结肠癌中的表达及其临床意义 被引量:2

Expressions of hypoxia inducible factor-lα and autophagy related protein Beclin-1 in colon cancer and their clinical significances

摘要 目的探讨缺氧诱导因子1α(HIF-1α)和自噬相关蛋白Beclin-l在结肠癌中的表达及其临床意义。方法选取2017年1月至2018年12月丹东市第一医院收治的120例结肠癌患者,采用免疫组织化学SABC法检测癌组织和相应癌旁组织中HIF-1α、BeClin-1蛋白的表达,并探讨其与结肠癌患者临床病理特征的关系。应用蛋白质印迹法检测20例新鲜结肠癌组织及其癌旁组织中HIF-1α和Beclin-1蛋白的表达。结果免疫组织化学结果显示,HIF-1α和Beclin-l在结肠癌组织中的阳性表达率均高于癌旁组织[75.0%(90/120)比21.7%(26/120),60.8%(73/120)比12.5%(15/120)],差异均有统计学意义(X^2值分别为68.343、60.359,均P<0.01)。蛋白质印迹法结果显示,20例新鲜结肠癌组织中HIF-1α、Beclin-l蛋白的相对表达量较癌旁组织中升高(1.98±0.66比0.76±0.55,1.50±0.40比0.46±0.35),差异均有统计学意义(t值分别为6.912、8.315,均P<0.01)。结肠癌组织中HIF-1α和Beclin-1的表达均与肿瘤TNM分期、分化程度、浸润深度、淋巴结转移和神经侵袭有关(HIF-1α:X^2值分别为9.074、15.215、10.101、11.610、9.979;Beclin-1:X^2值分别为11.285、4.858、24.436、9.354、4.632;均P<0.05),而与患者年龄、性别、肿瘤部位、肿瘤直径及脉管癌栓均无关(均P>0.05)。HIF-1α和Beclin-1蛋白在结肠癌组织中的表达呈正相关(r=0.483,P<0.01)。结论HIF-1α和Beclin-1蛋白在结肠癌组织中表达升高,两者相互作用共同参与了结肠癌的发生与发展。 Objective To investigate the expressions of hypoxia inducible factor-lα(HIF-lα)and autophagy related protein Beclin-1 in colon cancer and their clinical significances.Methods From January 2017 to December 2018,120 patients with colon cancer who were admitted to Dandong First Hospital were selected.The expressions of HIF-1αand Beclin-1 in colon cancer and corresponding adjacent tissues were detected by immunohistochemical SABC method.The relationship of the expressions of HIF-1αand Beclin-1 with the clinicopathological features of colon cancer patients was also discussed.The expressions of HIF-1αand Beclin-1 in 20 pairs of fresh colon cancer and paracancerous tissues were detected by Western blot.Results The results of immunohistochemistry showed that the positive expression rates of HIF-1αand Beclin-1 proteins in colon cancer tissues were significantly higher than those in paracancerous tissues[75.0%(90/120)vs.21.7%(26/120),60.8%(73/120)vs.12.5%(15/120)],and the differences were statistically significant(X^2 values were 68.343 and 60.359,both P<0.01).The results of Western blot showed that the expressions of HIF-1αand Beclin-1 proteins in 20 pairs of fresh colon cancer tissues were significantly higher than those in paracancerous tissues(1.98±0.66 vs.0.76±0.55,1.50±0.40 vs.0.46士0.35),and the differences were statistically significant(t values were 6.912 and 8.315,both P<0.01).The expressions of HIF-1αand Beclin-1 in colon cancer tissues were related to TNM stage,degree of differentiation,depth of infiltration,lymph node metastasis and nerve invasion(HIF-1α:X2 values were 9.074,15.215,10.101,11.610 and 9.979;Beclin-1:X^2 values were 11.285,4.858,24.436,9.354 and 4.632;all P<0.05),but not with age,sex,tumor location,tumor diameter and vascular tumor thrombus(all P>0.05).There was a positive correlation between the expressions of HIF-1α and Beclin-1 proteins in colon cancer(r=0.483,P<0.01).Conclusion The expressions of HIF-1α and Beclin-1 proteins in colon cancer tissues are significantly increased,and the interaction between them is involved in the occurrence and development of colon cancer.
