6株H9N2亚型禽流感病毒厦门分离株全基因序列测定与分析 被引量:5

摘要 为了解分离自厦门市养殖场、活禽交易市场和活禽屠宰场的H9N2亚型禽流感病毒(Avian influenza viruse,AIV)的遗传变异特征,本研究对分离到的6株H9N2亚型AIV进行全基因组序列测定、遗传进化分析和特殊位点的氨基酸分析。结果显示:6株分离株的HA裂解位点均符合低致病性禽流感病毒特征,HA受体结合位点尤其是234位氨基酸突变为L,表现出人流感病毒受体结合特性;NA存在颈部9个核苷酸缺失的高致病性分子特征;内部基因NS1基因发生P42S突变,M1基因发生N30D、T215A突变,M2基因发生S31N的突变,PB1-F2基因发生N66S突变,提示当前分离株已出现耐药性、致病性增强的变化。8个基因片段均属于欧亚谱系,内部基因与H7N9、H10N8、H10N6、H5N6亚型亲缘关系密切。本研究结果为本地区禽流感生物学特性研究和防控提供科学依据。 To master the molecular characteristics and genetic evolutionary relationship of the H9 N2 Avian influenza viruse(AIV)isolated from poultry farm,live-poultry market and live-poultry slaughtering factory of Xiamen,the complete genomes of six H9 N2 AIV isolates were amplified,sequenced and analyzed.The result showed that the six isolates were categorized as low pathogenic AIV,with typical HA cleavage site.The receptor binding site at site 234 was Leucine,which showed human influenza virus receptor binding characteristics.The six isolates had special high pathogenic NA sequence,which had 9 continuous amino acid deletion in the stalk region.There were mutations in the internal genes,including P42 S in NS1,both N30 D and T215 A in M1,S31 N in M2,N66 S in PB1-F2.These mutations indicated that the current isolates had a trend of increased resistance and increased pathogenicity.All genes of the six isolates belonged to The Durasian avian lineage and the six inner genes were closely related to H7 N9、H10 N8、H10 N6、H5 N6 subtype.The result will lay the foundation of biological characteristic study and provide scientific basis for prevention and control of AIV.
