摘要 目的优化反应条件,实现唾液淀粉酶(简称酶)活性的实时观察。方法比较超纯水和不同稀释倍数的pH6.8磷酸缓冲液(简称缓冲液)溶解的淀粉、酶量对酶活性的影响。于反应前加碘,比较优化反应条件前后淀粉褪色的速度。结果酶在缓冲液配制的淀粉中的活性高于纯水,配制淀粉的缓冲液浓度越低,酶活性越弱;酶活性随酶量增加,8滴酶最适合观察碘液褪色的过程。优化反应条件加速了反应前加碘的反应速度。结论本研究首次发现碘液抑制唾液淀粉酶活性,优化了酶促反应条件,实现了酶活性的实时观察,本研究将为影响唾液淀粉酶活性因素的实验教学带来极大便利。 Objective To realize the real-time observation of salivary amylase(SA) activity by optimizing reaction conditions. Methods The effect of starch dissolved in ultrapure water and pH 6.8 phosphate buffer saline(PBS) with different dilution ratio on enzyme activity was compared.The fading rate of iodine in starch solution before and after optimal reaction conditions was also compared.Results The activity of SA in starch dissolved in PBS was higher than that of pure water.The lower the concentration of buffer,the weaker the activity of SA.The SA activity increased with the amount of SA and 8 drops of SA was most suitable for observing the fading process of iodine.Optimized reaction conditions greatly accelerated the reaction velocity of SA.Conclusion In this study,iodine solution was found to be an ihhibitor of SA for the first time.This study also optimized the enzymatic reaction conditions and realized the real-time observation of the SA activity,which will bring great convenience to the experimental teaching of factors affecting the activity of SA.
