摘要
目的探讨胃癌患者血清的BRCA1相关环指结构域-1(BARD1)水平及其临床意义。方法 GEPIA数据库在线分析胃癌组织和正常组织的BARD1水平;实时定量PCR(QPCR)检测2014年1月至2017年12月本院收治的177例胃癌患者及173例健康体检者的血清BARD1水平,分析血清BARD1水平与胃癌临床病理特征的关系。根据随访资料分析血清BARD1水平与胃癌预后的关系。结果 GEPIA数据库在线分析显示408例胃癌组织BARD1水平的中位数为3.87,高于36例正常组织的1.36(P<0.05);QPCR检测结果显示,177例胃癌患者的血清BARD1水平为4.034±1.941,高于健康体检者的1.427±0.799(t=16.367,P<0.001)。血清BARD1水平与性别、年龄、肿瘤位置和浸润深度无关(P>0.05),而与肿瘤直径、分化程度、Bormann分型、淋巴结转移、远处转移和TNM分期有关(P<0.05)。BARD1低水平者的中位总生存期为39.0个月,长于高水平者的36.0个月(P<0.05),多因素分析显示血清BARD1水平是影响胃癌预后的独立因素(OR=3.145,95%CI:2.897~4.268,P=0.017)。结论胃癌患者组织和血清中BARD1表达上调,该指标的血清水平与胃癌的恶性程度和预后有关,提示BARD1可能与胃癌的发生发展有关。
Objective To investigate the level of BRCA1-associated ring domain protein 1(BARD1) in serum of patients with gastric cancer and its clinical significance. Methods The BARD1 levels in gastric cancer tissues and normal tissues were analyzed online by GEPIA database, and serum BARD1 levels of 177 gastric cancer patients and 173 healthy physical examinees in our hospital from January 2014 to December 2017 were detected by real-time quantitative PCR(QPCR). Relationship between serum BARD1 level and clinicopathological characteristics of gastric cancer was analyzed. According to the follow-up data, the relationship between the serum BARD1 level and prognosis of gastric cancer was analyzed. Results On-line analysis of GEPIA database showed that the median level of BARD1 in 408 cases of gastric cancer was 3.87, higher than 1.36 in 36 cases of normal tissues(P<0.05). The results of QPCR showed that the serum BARD1 level of 177 patients with gastric cancer was 4.034±1.941, higher than 1.427±0.799 of the healthy physical examinees(t=16.367, P<0.001). The serum level of BARD1 in gastric cancer was not related to gender, age, tumor location and invasion depth(P>0.05), but related to tumor diameter, differentiation degree, Bormann classification, lymph node metastasis, distant metastasis and TNM stage(P<0.05). The median overall survival was 39.0 months in the low-level group, longer than 36.0 months in high-level group(P<0.05). Multivariate analysis showed that serum BARD1 level was an independent factor influencing the prognosis of gastric cancer(OR=3.145, 95% CI: 2.897-4.268, P=0.017). Conclusion The expression of BARD1 is up-regulated in the tissues and serum of patients with gastric cancer. The serum level of BRDA1 is related to the malignant degree and prognosis of gastric cancer, suggesting that BARD1 may be related to the occurrence and development of gastric cancer.
