鹿茸多肽对单侧颈总动脉结扎所致轻度认知功能障碍模型大鼠的保护作用研究 被引量:6

摘要 目的通过观察鹿茸多肽对单侧颈总动脉结扎法复制的轻度认知功能障碍(Mild Cognitive Impairment,MCI)模型大鼠行为学指标、血清、海马组织中脑源性神经营养因子(Brain Derived Neurotrophic Factor,BDNF)、神经生长因子(Nerve Growth Factor,NGF)含量及表达水平的影响,探讨鹿茸多肽对模型大鼠的保护作用机制。方法大鼠60只,雌雄各半,随机分为假手术组、模型组、阳性对照组、鹿茸多肽高、中、低剂量组,给药前采用单侧颈总动脉结扎法复制MCI动物模型,给药周期18d,于末次给药2h后利用跳台实验检测各组模型大鼠行为学指标,利用酶联免疫法(ELISA)检测各组模型大鼠血清中BDNF、NGF含量,利用蛋白质印迹法(Western Blot)检测各组大鼠海马组织中BDNF、NGF表达水平。结果跳台实验结果表明,与模型组比较,阳性对照组、鹿茸多肽高、中、低剂量组逃避潜伏期明显降低(P<0.05或P<0.01),阳性对照组、鹿茸多肽高、中剂量组3min内错误次数明显减少(P<0.01);ELISA实验结果表明,与模型组比较,阳性对照组、鹿茸多肽高、中、低剂量组血清中BDNF含量明显增多(P<0.01),NGF含量明显增多(P<0.05或P<0.01);Western Blot实验结果表明,与模型组比较,阳性对照组、鹿茸多肽高、中、低剂量组海马组织中BDNF、NGF表达水平明显明显升高(P<0.05)。结论鹿茸多肽对单侧颈总动脉结扎法复制的MCI模型大鼠具有保护作用,作用机制可能与提升模型大鼠血清、海马组织中BDNF、NGF含量及表达水平相关。 OBJECTIVE To observe the effects of pilose antler polypeptide on behavioral indexes,serum and hippocampal BDNF,NGF content and expression levels in rats with mild cognitive impairment induced by unilateral common carotid artery ligation,and to explore the effect of velvet antler polypeptide on model rats.Protective mechanism.METHODS Sixty rats,half male and half female,were randomly divided into sham group,model group,positive control group,velvet antler polypeptide high,medium and low dose groups.The unilateral common carotid artery ligation was used to replicate mild cognitive function before administration.The animal model of the disorder was administered for 18 days.After 2 hours of the last administration,the behavioral indexes of the rats in each group were detected by the platform test.The serum levels of BDNF and NGF were detected by enzyme-linked immunosorbent assay(ELISA).The expression levels of BDNF and NGF in hippocampus of each group were detected by Western Blot.RESULTS TThe results of the platform test showed that compared with the model group,the escape latency of the high-,medium-,and low-dose groups of the positive control group and the antler polypeptide was significantly lower(P<0.05 or P<0.01),and the positive control group,the antler polypeptide,and the middle dose group were within 3 minutes.The number of errors was significantly reduced(P<0.01).ELISA showed that compared with the model group,the serum levels of BDNF in the high,middle and low dose groups of the positive control group and the antler polypeptide were significantly increased(P<0.01),NGF content increased significantly(P<0.05 or P<0.01);The results of Western Blot showed that compared with the model group,the expression levels of BDNF and NGF in the hippocampus of the positive control group and the antler polypeptide group were significantly increased(P<0.05).CONCLUSION The antler polypeptide has a protective effect on the mild cognitive impairment model rat with unilateral common carotid artery ligation.The mechanism may be related to the BDNF and NGF content and expression levels in the serum and hippocampus of the model rats.
