摘要
目的研究UBAP 1和LMP 1基因在鼻咽癌组织中的表达和相关机制。方法本研究采用免疫组化(immunohistochemistry)检测鼻咽癌细胞的增殖,通过Western blot和RT-PCR检测UBAP 1和LMP 1基因在人鼻咽癌发生发展过程中的表达。结果免疫组化结果表明,与正常CK相比,检测到UBAP 1表达明显下调,细胞增殖数目减少;而LMP 1则明显上调,导致其细胞增殖数目增加。NPC细胞中的IL-6和TNF-α的水平与对照相比有显著的增加;为了验证之前检测的UBAP 1和LMP 1基因表达的准确性,本研究检测了UBAP 1以及LMP 1蛋白在NPC细胞中的表达,结果显示与基因表达结果的趋势相一致。结论UBAP 1和LMP 1基因与癌细胞的增殖和迁移相关,且UBAP 1和LMP 1也可能具有不同的信号通路效应因子,是早期诊断和靶向治疗的潜在新靶点,也是NPC预后的新分子标记。
Objective To study the expressions and related mechanisms of UBAP 1 and LMP 1 genes in nasopharyngeal carcinoma.Methods In this study,immunohistochemistry was used to detect the proliferation of nasopharyngeal carcinoma cells,and the expressions of UBAP 1 and LMP 1 genes in human nasopharyngeal carcinoma were detected by western blot and RT-PCR.Results Immunohistochemistry showed that UBAP 1 expression was significantly down-regulated and the number of cell proliferation was decreased compared with normal CK,while LMP 1 was detected to have a significant up-regulation trend,which resulted in an increase in the number of cell proliferation.The levels of IL-6 and TNF-αin NPC cells were significantly increased compared with the control.In order to verify expression accuracy of previously detected UBAP 1 and LMP 1 genes,we examined the expressions of UBAP 1 and LMP 1 proteins in NPC cells.The results were consistent with the trend of gene expression results.Conclusion UBAP 1 and LMP 1 genes are involved in the proliferation and migration of cancer cells,which could function through different signaling pathways.Our results provides information on potential new targets for the early diagnosis and targeted therapy,and also novel prognostic molecular marker for NPC.
