摘要 为了稳定金丝楸增殖、增加组培苗有效新梢数、提高生根率,采用改变继代培养方式、将增殖和壮苗培养结合、添加活性炭等措施,以使金丝楸组培工厂化生产顺利进行.结果表明:选择春季萌发的枝条为外植体可以获得较高的诱导效率,培养基以WPM中附加0.8 mg/L BA、0.1 mg/L IBA为宜,诱导率为95%.增殖培养基以WPM附加2.0 mg/L BA、0.1 mg/L IBA为宜,增殖系数为3.3;增殖培养20 d后,形成大量丛生芽,不进行切割,直接转入MS+0.8 mg/L BA+0.1 mg/L IBA中进行壮苗培养,平均苗高2.6 cm,壮苗率达到78%.将苗高大于2.0 cm的单株在大量元素减半的MS中进行生根培养;培养基中添加活性炭可以促进根系发育,活性炭用量为0.5 g/L,生根率为95.2%,根数4~5条,根长1.2~2.0 cm. In order to stabilize the proliferation,increase the effective number of new shoots of tissue culture seedlings and improve the rooting rate,measures such as changing the secondary culture mode,combining the proliferation with the strong seedling culture,adding activated carbon and so on,were adopted to make the tissue culture factory production of Catalpa bungi var. go on smoothly.The results showed that high induction efficiency can be achieved by selecting the spring germinating branches as explants,and adding BA 0.8 mg/L and IBA 0.1 mg/L to the WPM is appropriate,and the induction rate is 95%.The proliferation medium should be added with WPM BA 2.0 mg/L and IBA 0.1 mg/L,and the proliferation coefficient was 3.3.After proliferation and culture for 20 days,a large number of clumps of buds were formed,without cutting,and directly transferred into MS +BA 0.8 mg/L + IBA 0.1 mg/L for strong seedling culture.The average height of the seedlings was 2.6 cm,and the strong seedling rate reached 78%.The single plant with seedling height of 2.0 cm was rooted and cultured in MS with a large number of elements halved.The addition of activated carbon in the medium can promote the root development.The dosage of activated carbon is0.5 g/L,the rooting rate is 95.2%,the number of roots is 4~5,and the length of roots is 1.2~2.0 cm.
