应用流式细胞术及IDT307探究血小板5-羟色胺转运体功能

摘要 目的:应用流式细胞术测定血小板转运IDT307的方法来测定并比较不同人群血小板5-羟色胺转运体的功能差异。方法:选取34例汕头大学精神卫生中心门诊及汕头市中心医院心理门诊的患者以及性别年龄相匹配的25例健康对照者,使用流式细胞仪测定外周血小板5-羟色胺转运体转运IDT307的阳性率及荧光强度值。结果:各组血小板转运IDT307的阳性率比较差异无统计学意义(P>0.05)。患者组血小板转运IDT307入细胞内的荧光强度值为110.2±28.5,健康对照组为83.3±19.9,二者比较差异有统计学意义(P<0.01)。患者组血小板5-羟色胺转运体的转运能力显著高于健康对照组。男、女患者血小板转运IDT307入细胞内的荧光强度值差异无统计学意义(P>0.05);抑郁、焦虑、强迫障碍患者血小板转运IDT307入细胞内的荧光强度值比较差异也无统计学意义(P>0.05)。结论:抑郁、焦虑、强迫障碍患者血小板5-羟色胺转运体功能都升高,并且不受性别与疾病状态的影响。应用流式细胞术测定血小板转运IDT307的方法来测定血小板5-羟色胺转运体的功能,检测结果稳定可靠。 Objective: Flow cytometry was used to measure platelet transporter IDT307 and compare the functional differences of platelet 5-hydroxytryptamine(5-HT) transporter in different population. Methods:34 outpatients from the Shantou University Mental Health Center and the Psychological Clinic of Shantou Central Hospital and 25 healthy controls matched by gender and age were selected to measure the positive rate and fluorescence intensity of peripheral platelet 5-HT transporter IDT307 by flow cytometry. Results:There was no significant difference in the positive rate of platelet transporting IDT307 between groups(P>0.05);the fluorescence intensity of platelet transporting IDT307 into the cells in the patient group was 110.2±28.5,and that in the healthy group was 83.3 ± 19.9(P<0.01). The transport capacity of 5-HT transporter in patients was significantly higher than that in healthy controls. There was no significant difference in the fluorescence intensity of platelet transport IDT307 between male and female patients(P>0.05);there was no significant difference in the fluorescence intensity of platelet transport IDT307 between depression,anxiety and obsessivecompulsive disorder patients(P>0.05). Conclusion:The function of platelet 5-HT transporter was increased in patients with depression, anxiety and obsessive-compulsive disorder, and was not affected by gender and disease status. The method using IDT307 to observe the transporting function of human platelets is stable and reliable.
