天南星水提取物对胃癌大鼠细胞中PKM2、mTOR基因表达的影响 被引量:9

Effect of Water Extract of Rhizoma Arisaema on the Expression of PKM2 and mTOR Genes in Gastric Cancer Rat Cells

摘要 探究天南星水提取物对胃癌大鼠细胞中PKM2、mTOR表达的影响。选取50只SD雄性大鼠,10只作为正常组,其余40只建立胃癌模型,分为胃癌组、药物对照组、低、高浓度组,并分别进行干预。观察胃癌大鼠癌组织细胞增殖、凋亡情况,检测各组大鼠胃动素、胃泌素水平、PKM2、mTOR、Bcl-2、PI3K、Akt表达。研究结果显示,高浓度组大鼠胃动素、胃泌素水平为137.65 pg/mL、88.76 pg/mL,凋亡率为49.73%,均高于胃癌组、药物对照组、低浓度组(p<0.05)。高浓度组大鼠增殖率为7.55%,PKM2、mTOR、Bcl-2、PI3K、Akt相对表达量分别为1.37、1.35、1.37、1.42、1.39,均低于胃癌组、药物对照组、低浓度组(p<0.05)。实验结果表明,在天南星水提取物的干预下,胃癌大鼠胃功能得到改善,癌组织细胞增殖率下降,凋亡率上升,PKM2、mTOR、Bcl-2、PI3K、Akt表达受到明显的调控,其能力呈现浓度依赖,说明天南星水提取物能够抑制胃癌细胞增殖,促进胃癌细胞凋亡,为胃癌的临床治疗提供一定的理论依据。 To investigate the effect of water extract of Rhizoma Arisaema on the expression of PKM2 and mTOR in gastric cancer rat cells. 50 SD male rats were selected, 10 of which were used as the normal group, and the remaining 40 were used to establish the gastric cancer model. The rats were divided into gastric cancer group, drug control group, low and high concentration group, and were treated with intervention. The proliferation and apoptosis of cancer cells in gastric cancer rats were observed, and the expressions of motilin, gastrin, PKM2, mTOR, bcl-2, PI3 K and Akt in each group were detected. The results showed that the levels of motilin and gastrin in the high-concentration group were 137.65 pg/ml and 88.76 pg/ml, respectively, and the apoptosis rate was 49.73%, which were all higher than those in the gastric cancer group, drug control group and low-concentration group(p<0.05). The proliferation rate of rats in the high-concentration group was 7.55%, and the relative expression levels of PKM2, mTOR, bcl-2, PI3 K and Akt were 1.37, 1.35, 1.37, 1.42 and 1.39, respectively, which were lower than those in the gastric cancer group, drug control group and low-concentration group(p<0.05). The results showed that the gastric function of gastric cancer rats was improved, the proliferation rate of cancer cells was decreased and the apoptosis rate was increased. The expression of PKM2, mTOR, Bcl-2, PI3 K and Akt was obviously regulated by the water extract of rhizoma Arisaema. The anticancer ability of the water extract of Rhizoma Arisaema was concentration-dependent, indicating that the water extract of Rhizoma Arisaema could inhibit the proliferation of gastric cancer cells, promote apoptosis of gastric cancer cells and provide a theoretical basis for clinical treatment of gastric cancer.
