摘要
目的探讨番茄红素对肾脏缺血再灌注损伤的防护作用及其对自噬机制的影响。方法将SD大鼠随机分为假手术组(A组)、缺血再灌注组(B组)、缺血再灌注+番茄红素组(C组)、缺血再灌注+番茄红素+3-MA组(D组)。比较4组大鼠肾组织丙二醛(MDA)、超氧化物歧化酶(SOD)、Na^+,K^+-ATP酶、Ca2^+/ATP酶的变化,采用HE染色比较4组大鼠肾脏病理学改变,采用免疫印迹法检测自噬相关基因微管相关蛋白轻链3(LC3)的表达变化。结果与A组相比,B组大鼠肾小管上皮细胞呈明显缺血性改变,其MDA升高,SOD、Na^+,K^+-ATP酶、Ca2^+/ATP酶活性降低;与B组相比,C组大鼠肾小管上皮细胞缺血性改变减轻,MDA明显降低,SOD、Na^+,K^+-ATP酶、Ca2^+/ATP酶活性增高;番茄红素可促进肾脏LC3Ⅱ的表达,且番茄红素的上述防护作用可被3-MA抑制。结论番茄红素可能通过激活自噬来减轻肾脏缺血再灌注损伤。
Objective To investigate the protective effect of lycopene on acute renal ischemia reperfusion injury and its effect on autophagy mechanism. Methods SD rats were randomly divided into sham operated group( group A);ischemia reperfusion group( group B);ischemia reperfusion + lycopene injection group( group C);ischemia reperfusion + lycopene + 3-MA injection group( group D). The changes of malondialdehyde in renal tissue( MDA),superoxide dismutase( SOD),Na^+,K^+-ATPase、Ca2^+/ATPase in kidney were compared. HE staining was used to compare the pathological changes in the kidneys of the four groups,and the expression of autophagy-related genes microtubule-associated protein light chain 3( LC3) was detected by immunoblotting. Results Compared with group A,there were some ischemic changes in the renal tubular epithelial cells,the content of MDA increased significantly and SOD,Na^+,K^+-ATPase,Ca2^+/ATPase decreased sharply in group B. Lycopene rescued the renal ischemia/reperfusion injury,and this could be inhibited by autophagy inhibitor 3-methyladenine( 3-MA). Also,lycopene promoted the expression of LC3 Ⅱ in kidney. Conclusion Lycopene may alleviate renal ischemia reperfusion injury by activating autophagy.
