摘要
目的探讨细胞色素P450c17a(CYP17A1)和缺氧诱导因子-1α(HIF-1α)在脑胶质瘤组织中的表达情况及与患者临床特征的关系。方法选择88例脑胶质瘤患者的脑胶质瘤组织和44例外伤后实施开颅手术且术后病理学检查无病变患者的正常脑组织,采用免疫组织化学染色法检测HIF-1α和CYP17A1蛋白的表达情况,分析HIF-1α和CYP17A1蛋白表达与脑胶质瘤患者年龄、性别、世界卫生组织(WHO)胶质瘤分级及肿瘤直径的关系。结果脑胶质瘤组织中HIF-1α和CYP17A1蛋白的阳性表达率分别为67.05%和76.14%,均明显高于正常脑组织的6.82%和11.36%,差异均有统计学意义(P﹤0.01)。WHO分级为Ⅲ~Ⅳ级的脑胶质瘤患者脑胶质瘤组织中HIF-1α和CYP17A1蛋白的阳性表达率均高于WHO分级为Ⅰ~Ⅱ级的脑胶质瘤患者,差异均有统计学意义(P﹤0.05);不同年龄、性别、肿瘤直径的脑胶质瘤患者脑胶质瘤组织中HIF-1α和CYP17A1蛋白的阳性表达率比较,差异均无统计学意义(P﹥0.05)。结论脑胶质瘤组织中HIF-1α和CYP17A1蛋白的阳性表达率较高,且CYP17A1和HIF-1α蛋白表达情况与脑胶质瘤患者的WHO分级密切相关。
Objective To investigate the relationship between the expression of cytochrome P450c17a(CYP17A1),hypoxia-inducible factor-1α(HIF-1α)and the clinical characteristics in patients with glioma.Method Eighty-eight tissue specimens from patients with glioma were collected and 44 specimens of normal brain tissues were obtained from another cohort of patient after craniotomy.Immunohistochemical staining was used to detect HIF-1αand CYP17A1 protein expression.The relationship of HIF-1α,CYP17A1 protein and age,gender,World Health Organization(WHO)classification of gliomas,and tumor diameter was investigated.Result The positive expression rates of HIF-1αand CYP17A1 protein in glioma tissues were 67.05%and 76.14%,which were significantly higher than the 6.82%and 11.36%in normal brain tissues(P<0.01).The positive expression rate of HIF-1αand CYP17A1 protein in glioma tissues of WHO grade III to IV was significantly higher compared to that in those with WHO grade I to II(P<0.05);the positive expression rates of HIF-1αand CYP17A1 protein were independent of age,gender and tumor diameter among glioma patients(P>0.05).Conclusion The positive expression rates of HIF-1αand CYP17A1 proteins in glioma tissues is significantly increased,and closely relate to the WHO classification of glioma patients.
