摘要
目的探讨长链非编码RNA(lncRNA)LINC00968在非小细胞肺癌(NSCLC)中的表达及其临床意义。方法使用实时定量反转录聚合酶链反应(qRT-PCR)法检测85例NSCLC患者肿瘤组织及对应癌旁组织中lncRNA LINC00968的表达水平,并分析肿瘤组织中lncRNA LINC00968表达水平与NSCLC患者临床特征的关系。应用qRT-PCR法检测转染与未转染细胞中lncRNA LINC00968的表达水平,使用噻唑蓝(MTT)法检测转染细胞的增殖活性,使用蛋白质印迹法(Western blot)及qRT-PCR法检测lncRNA LINC00968表达对表皮生长因子受体(EGFR)蛋白及mRNA表达的影响。结果NSCLC患者肿瘤组织中lncRNA LINC00968的相对表达量低于癌旁组织(P﹤0.05)。不同组织分化程度、临床分期、淋巴结转移情况NSCLC患者肿瘤组织中lncRNA LINC00968的相对表达量比较,差异均有统计学意义(P﹤0.05)。转染48、72 h后,转染lncRNA LINC00968表达质粒的A549细胞的增殖能力低于转染空载体的A549细胞(P﹤0.05)。转染lncRNA LINC00968表达质粒的A549细胞中EGFR蛋白和mRNA的相对表达量均低于转染空载体的A549细胞(P﹤0.05)。结论lncRNA LINC00968在NSCLC患者肿瘤组织中表达下调,并可影响EGFR蛋白表达,lncRNA LINC00968表达水平与NSCLC的发生发展有关。
Objective To investigate the expression of long noncoding RNA(lncRNA)LINC00968 in non-small cell lung cancer(NSCLC)and its clinical significance.Method The expression level of lncRNA LINC00968 in tumor tissues and adjacent tissues of 85 NSCLC patients was detected by real-time quantitative reverse transcription polymerase chain reaction(qRT-PCR),besides,the relationship between the expression level of lncRNA LINC00968 in tumor tissues and the clinical characteristics of NSCLC patients was analyzed.The expression level of lncRNA LINC00968 in transfected and non-transfected cells was detected by qRT-PCR;MTT assay was used to detect the proliferative activity of the transfected cells;Western blot and qRT-PCR were applied to determine the effect of lncRNA LINC00968 on the expression of epidermal growth factor receptor(EGFR)protein and mRNA.Result The relative expression of lncRNA LINC00968 in NSCLC tissues was lower than that in the adjacent tissues(P<0.05).There was significant difference in the relative expression of lncRNA LINC00968 in NSCLC tissues of NSCLC patients with different degree of tissue differentiation,clinical stage and lymph node metastasis(P<0.05).After 48 and 72 hours of transfection,the proliferative ability of lncRNA LINC00968-transfected group was significantly lower compared to cells transfected with empty vector(P<0.05).Furthermore,the expression level of EGFR protein and mRNA in lncRNA LINC00968-transfected A549 cells were significantly lower than that in the empty vector transfection A549 cells(P<0.05).Conclusion The expression of lncRNA LINC00968 is down-regulated in tumor tissues of NSCLC patients and may affect the expression of EGFR.The expression level of lncRNA LINC00968 is related to the occurrence and development of NSCLC.
