摘要 目的探究芦荟大黄素(aloe emodin,AE)对便秘小鼠结肠肌电表达的影响.方法30只小鼠均匀分布,分对照组、模型组、AE组.用复方地芬诺酯片混悬液灌胃法建立慢性传输型便秘模型,AE治疗AE组.检测小鼠每日排便量、粪便性状和结肠肌电活动,从而探讨AE对便秘小鼠结肠肌电表达的影响,同时用免疫组织化学方法测定各组小鼠结肠P物质(P substance,SP)和血管活性肠肽(vasoactive intestinal peptide,VIP)的表达情况.结果三组小鼠的结肠慢波表现与正弦波样曲线相接近.模型组结肠慢波频率为(47.97±2.63)次/min,与对照组(43.75±0.69)次/min相比明显加快,差异有高度统计学意义(P<0.01),AE组结肠慢波频率与模型组比较明显减慢,差异有高度统计学意义(P<0.01);模型组结肠慢波频率变异系数(6.02±0.23)%,与对照组(1.83±0.38)%相比明显增大(P<0.01),AE组小鼠频率变异系数(2.77±0.19)%,与模型组相比更趋于正常(P<0.01);模型组结肠慢波振幅(0.05±0.03)mV,与对照组(0.16±0.03)mV相比显著减少(P<0.01),AE组振幅(0.13±0.03)mV,与模型组相比明显回升(P<0.01);模型组结肠慢波振幅变异系数(62.79±1.71)%,与对照组(10.13±0.55)%相比明显增大(P<0.01),AE组小鼠振幅变异系数(25.31±1.97)%,与模型组相比明显减小(P<0.01).免疫组化实验结果中小鼠肠道SP和VIP含量较对照组明显升高(P<0.01).结论AE对便秘小鼠具有缓解性作用,其缓解机制可能是通过升高SP和VIP来实现的. Objective To investigate the effect of aloe-emodin on colonic myoelectrical expression in mice with constipation.Methods 30 mice were evenly divided into three groups at random,the control group,the model group and the AE group.A model of chronic transmission constipation was established by intragastrically administered with a compound Diphenoxylate tablets and aloe-emodin was used to treat the AE group.The daily bowel movements,faecal character and myoelectric activity of the mice were measured to investigate the effect of aloe-emodin on colon myoelectrical expression in mice with constipation.Expressions of SP(P substance)and VIP(vasoactive intestinal peptide)in mice colon in three groups were detected by immunohistochemical method.Results The colonic slow waves of the three groups of mice appeared to be similar to that of a sine wave-like curve.Compared with the control group(43.75±0.69 times/min),the frequency of colonic slow waves(47.97±2.63 times/min)in the model group was faster,and the difference is of statistical significance(P<0.01).Compared with the model group,the frequency of colonic slow waves was slow in the AE group and the difference showed statistical significance(P<0.01).Compared with the control group(1.83±0.38)%,the frequency coefficient of variation of the model group was significantly larger(6.02±0.23)%and difference was of statistical significance(P<0.01).Compared with the model group,the coefficient of variation of the frequency of colonic slow waves in the AE group was closer to the normal level and the difference was of statistical significance(P<0.01).Compared with the control group(0.16±0.03)mV,the amplitude of colonic slow wave of the model group(0.05±0.03)mV was significantly lessened,and the difference was statistical significant(P<0.01).Compared with the model group,the amplitude of the AE group was(0.13±0.03)mV increased significantly(P<0.01).Compared with the control group(10.13±0.55)%,the coefficient variation(62.79±1.71)%of the amplitude of the colonic slow wave in mice with constipation in the model group increased significantly and the difference was of statistical significance(P<0.01).Compared with the model group,the coefficient variation(25.31±1.97)%of slow-wave vibration in the AE group was significantly lower and the difference was of statistically significance(P<0.01).The content of SP and VIP in intestinal tract of mice was significantly higher than that of control group in immunohistochemical results(P<0.01).Conclusion Aloe-emodin has a palliative effect on constipation mice and its mitigation mechanism may be achieved by elevating SP and VIP.
