抑制白细胞介素IL-1β后脊髓钝挫伤大鼠运动功能恢复的研究

摘要 目的本实验将研究IL-1β对大鼠脊髓钝挫伤后运动功能的影响,在最大程度保留神经功能的前提下,在临床上对脊髓神经病变提供治疗依据。方法建立脊髓损伤动物模型,BBB评分评价大鼠SCI后后肢的运动功能;采用RT-PCR、WesternBlot、免疫组化对IL-1β基因和蛋白质表达进行定量和定位分析。结果(1)通过BBB评分分析得出IL-1β基因干扰后,大鼠的运动恢复能力较SCI组和SCI+生理盐水组显著提高;(2)SCI+IL-1βRNAi组的IL-1β基因的mRNA的表达显著低于SCI组和SCI+生理盐水组;(3)SCI+IL-1βRNAi组的IL-1β基因的蛋白的表达显著低于SCI组和SCI+生理盐水组;(4)免疫组化实验结果显示组织中SCI+IL-1βRNAi组的IL-1β基因的表达显著低于SCI组和SCI+生理盐水组。结论IL-1β基因被siRNA干扰后,大鼠的运动恢复能力较不做处理的脊髓损伤大鼠有所提高,且提高效果显著,有统计学差异。 Objective:To investigate the effects of IL-1βon motor function after spinal cord contusion in rats,and provide clinical evidence for spinal neuropathy on the premise of maximally retaining neurological function.Methods:An animal model of spinal cord injury.The BBB score was used to evaluate the motor function of hind limbs after SCI in rats.RT-PCR,Western Blot,immunohistochemistry was used to quantify the expression of IL-1βgene and protein.Results:1.After the interference of IL-1βgene by BBB score analysis,the exercise recovery ability of rats was significantly higher than that of SCI group and SCI+saline group.2.The mRNA expression of IL-1βgene in SCI+IL-1βRNAi group was significantly lower than that in SCI group and SCI+saline group.3.The expression of IL-1βgene protein in SCI+IL-1βRNAi group was significantly lower than that in SCI group and SCI+saline group.4 Immunohistochemistry resμLts showed that the expression of IL-1βgene in SCI+IL-1βRNAi group was significantly lower than that in SCI group and SCI+saline group.Conclusion:After the IL-1βgene was interfered by siRNA,the exercise recovery ability of rats was higher than that of the spinal cord injury rats,and the improvement effect was statistically significant.
