卵巢癌中SUMO1P3表达及对细胞生物学行为的影响 被引量:2

摘要 目的探讨长链非编码SUMO1P3在卵巢癌细胞中的表达及对卵巢癌生物学行为的影响。方法采用实时荧光定量PCR(QPCR)检测上皮卵巢癌细胞系HO8910、SKOV3和CAOV3和正常卵巢上皮细胞系IOSE80中SUMO1P3表达;si-SUMO1P3转染CAOV3细胞以沉默SUMO1P3表达(si-SUMO1P3组),另转染阴性对照质粒为对照组(si-NC组);MTT法、划痕实验和Transwell实验分别检测细胞增殖、迁移和侵袭能力的变化;Western blotting检测E-cadherin、Vimentin、β-catenin、Cyclin D1和c-Myc蛋白的表达。结果卵巢癌HO8910、SKOV3和CAOV3细胞系中SUMO1P3表达水平分别为3.42±0.21、3.61±0.32和3.77±0.35,均高于正常卵巢上皮细胞系IOSE80的0.96±0.023,差异具有统计学意义(P<0.05)。与si-NC组的0.97±0.02比较,si-SUMO1P3组SUMO1P3表达明显降低(0.37±0.10,P<0.05)。MTT结果显示,si-SUMO1P3组24、48、72、96 h细胞增殖率分别为(89.9±17.1)%、(177.4±30.7)%、(364.6±38.8)%、(470.3±59.9)%,72和96 h细胞增殖率明显低于si-NC组。si-SUMO1P3组细胞24h迁移率为(37.8±5.8)%,显著低于si-NC组的(75.8±10.3)%;si-SUMO1P3组细胞侵袭数为159±8,显著低于si-NC组的248±9,差异均有统计学意义(P<0.05)。与si-NC组比较,si-SUMO1P3组Vimentin、β-catenin、Cyclin D1和c-Myc表达降低,E-cadherin表达增加。结论沉默SUMO1P3表达通过抑制β-catenin信号途径抑制上皮卵巢癌细胞的增殖、侵袭和迁移。 Objective To investigate the expression of long-chain noncoding SUMO1P3 in ovarian cancer and its effect on the biological behavior of ovarian cancer.Methods The expression of SUMO1P3 in epithelial ovarian cancer cell lines HO8910,SKOV3 and CAOV3 and normal human ovarian epithelial cell line IOSE80 was detected by real-time fluorescence quantitative PCR(QPCR).si-SUMO1P3 was transfected into CAOV3 cells to silence the expression of SUMO1P3(si-SUMO1P3 group),and the negative control plasmid was transfected into the control group(si-NC group);MTT assay,scratch assay and Transwell assay were used to detect cell proliferation,migration and invasion.The expression of E-cadherin,Vimentin,β-catenin,Cyclin D1 and c-Myc was detected by Western blotting.Results The expression of SUMO1P3 in HO8910,SKOV3 and CAOV3 cell lines was 3.42±0.21,3.61±0.32 and 3.77±0.35,respectively,which were higher than 0.96±0.023 in IOSE80 cell line(P<0.05).Compared with si-NC group(0.97±0.02),the expression of SUMO1P3 in si-SUMO1P3 group decreased significantly(0.37±0.10,P<0.05).MTT results showed that the proliferation rates of CAOV3 cells in si-SUMO1P3 group was(89.9±17.1)%,(177.4±30.7)%,(364.6±38.8)%,(470.3±59.9)%at 24,48,72 and 96 h,respectively,which was significantly lower than that in si-NC group.The number of cell invasion in si-SUMO1P3 group was 159±8,which was significantly lower than that in si-NC group(248±9),the difference was statistically significant(P<0.05).Compared with the si-NC group,the expression of Vimentin,β-catenin,Cyclin D1 and c-Myc in si-SUMO1P3 group decreased,while the expression of E-cadherin increased.Conclusion Silencing SUMO1P3 expression inhibits the proliferation,invasion and migration of epithelial ovarian cancer cells by inhibitingβ-catenin signaling pathway.
