摘要
目的研究miR-144在骨质疏松患者护理过程中的表达变化情况及其对骨质疏松的作用。方法采用实时定量聚合酶链式反应(real time qPCR)方法检测miR-144在骨质疏松患者中的表达及其在接受护理干预过程中的表达变化;采用CCK-8实验检测miR-144对人骨髓间充质干细胞(BMSCs)活性的影响;采用碱性磷酸酶(ALP)染色检测miR-144对人BMSCs成骨分化能力的影响。结果与正常对照组相比,骨质疏松患者血清中miR-144的表达显著降低;骨质疏松患者接受护理干预过程中,患者血清中miR-144的表达逐渐增高;CCK-8实验结果表明,miR-144 mimic显著提高人BMSCs活性,而miR-144 inhibitor显著抑制人BMSCs活性;ALP染色结果表明,miR-144 mimic显著促进人BMSCs向成骨方向分化,而miR-144 inhibitor显著抑制人BMSCs向成骨方向分化。结论miR-144在骨质疏松患者中表达降低,护理干预后miR-144的表达显著升高,且其能够调控人BMSCs活性和成骨分化能力。
Objective To investigate the expression changes of miR-144 in the nursing process of osteoporosis patients and its effect on osteoporosis.Methods Real-time quantitative polymerase chain reaction(real time qPCR)was used to detect the expression of miR-144 in osteoporosis patients and its expression changes during the course of nursing intervention.The CCR-8 assay was used to detect the effect of miR-144 on the cell viability of human bone marrow mesenchymal stem cells(BMSCs).Alkaline phosphatase(ALP)staining was used to detect the effect of miR-144 on the osteogenic differentiation of human BMSCs.Results Compared with the normal control group,the expression of miR-144 in the serum of patients with osteoporosis was significantly decreased.The expression of miR-144 in the serum of patients with osteoporosis was gradually increased during the intervention.The results of CCK-8 showed that miR-144 mimic significantly increased the activity of human BMSCs,while miR-144 inhibitor significantly inhibited the activity of human BMSCs.ALP staining showed that miR-144 mimic significantly promoted the differentiation of human BMSCs into osteogenic direction,while miR-144 inhibitor significantly inhibited the osteogenic differentiation of human BMSCs.Conclusion The expression of miR-144 is decreased in patients with osteoporosis.The expression of miR-144 is significantly increased after nursing intervention,and it can regulate the activity of human BMSCs and the ability of osteogenic differentiation.
