凝结芽孢杆菌对断奶仔猪肠道抗氧化和抗病毒能力的影响 被引量:3

Effect of Bacillus coagulans on intestinal antioxidant and antiviral ability in weaned piglets

摘要 为了研究日粮中添加凝结芽孢杆菌对断奶仔猪肠道抗氧化和抗病毒能力的影响,试验选取24头21日龄的健康断奶仔猪(杜×长×大),随机分为3组(对照组、BC6组、BC7组),每组8个重复,对照组饲喂基础日粮,BC6组饲喂基础日粮+凝结芽孢杆菌(2×106 cfu/g饲料),BC7组饲喂基础日粮+凝结芽孢杆菌(2×107 cfu/g饲料),于试验第21天屠宰取样,检测空肠黏膜组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)的活力和丙二醛(MDA)、过氧化氢(H2O2)含量,以及空肠炎性抗病毒和免疫调节相关基因[热休克蛋白1(HSPH1)、趋化因子(CCL2)、干扰素-α(IFN-α)、干扰素-β(IFN-β)、白细胞介素-4(IL-4)、抗氧化应激转录因子2(Nrf-2)基因]及相关蛋白[2’-5’寡聚腺苷酸合成酶1(OAS1)、抗黏液病毒1(MX1)]的相对表达量。结果表明:与对照组相比,BC6组空肠MDA、H2O2的含量显著降低(P<0.05),HSPH1、IFN-β基因的相对表达量显著下调(P<0.05),OAS1蛋白相对表达量显著升高(P<0.05);BC7组空肠H2O2的含量显著降低(P<0.05),SOD和CAT活力显著升高(P<0.05),IL-4、HSPH1和Nrf-2基因的相对表达量显著下调(P<0.05),IFN-β基因的相对表达量显著上调(P<0.05),MX1蛋白相对表达量显著升高(P<0.05)。说明日粮中添加凝结芽孢杆菌可以提高断奶仔猪肠道抗氧化能力和抗病毒能力,促进仔猪肠道吸收,从而维护肠道稳态,且日粮中添加2×107cfu/g的凝结芽孢杆菌效果更好。 The aim of this study was to investigate the effects of the addition of Bacillus coagulans in the diet on the intestinal antioxidant and antiviral capacity of weaned piglets.Twenty-four 21-day-old healthy weaned piglets(Duroc×Landrace×Yorkshire)were randomly divided into 3 groups with 8 replicates in each group.The three treatment groups were control group,BC6 group,and BC7 group.The control group was fed a basal diet,the BC6 group was fed a basal diet+Bacillus coagulans(2×106 cfu/g feed),and the BC7 group was fed a basal diet+Bacillus coagulans(2×107 cfu/g feed).Samples were taken at the 21 st day of the test to detect the content of SOD,CAT,MDA,H2O2 in jejunum mucosa,and to detect the relative expression of antiviral and immune related genes HSPH1,IFN-β,CCL2,IFN-α,IL-4,Nrf-2 and related proteins OAS1,MX1 in jejunum.The results showed that compared with the control group,the contents of MDA and H2O2 in jejunum of BC6 group were significantly decreased(P<0.05),the relative expression of HSPH1 and IFN-βgenes were significantly down-regulated(P<0.05),and the relative expression of OAS1 protein was significantly up-regulated(P<0.05).The amount of jejunum H2O2 of BC 7 group was significantly decreased(P<0.05),SOD and CAT activities were significantly increased(P<0.05),IL-4,HSPH1 and Nrf-2 genes were decreased(P<0.05),the relative expression of IFN-βgene was significantly up-regulated(P<0.05),and the relative expression of MX1 protein was significantly increased(P<0.05).The above results indicated that the addition of Bacillus coagulans in the diet can improve the intestinal antioxidant capacity and antiviral ability of weaned piglets,promote the intestinal absorption of piglets,thereby maintain intestinal homeostasis.Adding 2×107 cfu/g of Bacillus coagulans to the diet had a better effect.
