摘要
前期将1.4μg/mL浓度的多菌灵作为鉴别剂量对田间禾谷镰孢菌菌株进行敏感性测定,筛选出5株对多菌灵不敏感菌株,分别为W11-9、W11-3、J22-6、Q13-2、S28-9,选取对多菌灵表现敏感的2株禾谷镰孢菌菌株J22-2和W11-4,本研究以该7株菌株为研究材料,进行基因组提取并PCR扩增,通过比对分析其β微管蛋白基因(TUB 1、TUB 2)序列,从而验证生物学表型差异。通过比对发现它们之间TUB 1序列高度一致;J22-2和J22-6的TUB 2的1407位碱基由C变成A,但氨基酸种类没有改变,其余5株TUB 2序列并无差异。
In the previous research,1.4μg/mL carbendazim was used as the differential dose to determine the sensitivity of F.graminearum,and 5 strains of carbendazim-insensitive strains were selected,including W11-9,W11-3,J22-6,Q13-2,S28-9 and selected 2 strains of F.graminearum J22-2 and W11-4 which were sensitive to carbendazim.In this study,the 7 strains were used as materials for genome extraction and PCR amplification,which was performed to verity the difference in biological phenotype by analyzing the sequence of theβ-tubulin gene(TUB 1、TUB2).The results showed that there were no differences in the TUB 1 sequences of the seven strains.The 1407 base of the TUB 2 gene of J22-2 and J22-6 changed from C to A,but the amino acid type did not change,there were no differences in the other 5 strains of theβ2-tubulin gene.
