摘要
采用酵母浸粉、蛋白胨、琼脂粉等材料在特定的条件下培养枯草芽孢杆菌,使其分泌枯草芽孢杆菌酶,包括胞外酶和胞内酶;测试酶的导电能力及还原能力,结果表明:枯草芽孢杆菌胞内酶具有还原能力;当电源外加电压调至15 V时,胞内酶还原电位为867 mV,超过靛蓝所需要的还原电位-760 mV,枯草芽孢杆菌胞内酶可用做靛蓝电化学染色体系的还原剂,将其用于靛蓝电化学还原染色,发现当酶体系温度为60℃,pH为11,还原时间为40 min时,对靛蓝染料的还原效果较佳.
Bacillus subtilis is cultured with yeast extract,peptone and agar powder under specific conditions to secrete Bacillus subtilis enzymes.It produces extracellular enzymes and intracellular enzymes during culture.The conductivity and reductive capacity of the enzymes are tested.The results show that the reductive enzymes are intracellular enzymes of Bacillus subtilis,which are electrochemically stained with intracellular enzymes.When the applied voltage of the system is adjusted to 15 V,the reduction potential is 867 mV,which is higher than the reduction potential 760 mV required for indigo.It can be used as a reductant for indigo electrochemical chromosome system.When the temperature of the enzyme system is 60℃,the pH is 11,and the reduction time is 40 minutes,the reductive effect of indigo dye is the best.
