摘要
目的观察光遗传技术对帕金森病(PD)的治疗效果。方法将健康成年雄性Sprague Dawley大鼠随机分为实验组、病毒组、模型组和空白组,每组20只。模型组和实验组大鼠分别在右侧黑质致密部(SNc)和中脑腹侧被盖区(VTA)注射鱼藤酮建立单侧完全损毁的PD大鼠模型,实验组大鼠造模成功后注射腺相关病毒,病毒组大鼠不建立PD模型,仅注射腺相关病毒,空白组大鼠不给予任何干预措施。然后,各组大鼠在SNc植入光电极进行光刺激,采集各组大鼠光刺激前、中、后的脑电信号,采用LZ复杂度分析脑电信号的变化。结果PD大鼠与正常大鼠脑电信号各波段LZ复杂度与比较差异较大,光遗传学技术对导入病毒的实验组和病毒组大鼠脑电信号有较明显的影响,对未导入病毒的模型组和空白组大鼠脑电信号无明显影响;实验组大鼠光刺激时脑电信号θ波更趋近于空白组大鼠。结论光遗传技术对PD大鼠有治疗作用。
Objective To observe the therapeutic effect of optogenetics on Parkinson′s disease(PD)rats.Methods Healthy adult male Sprague Dawley rats were randomly divided into experiment group,virus group,model group and blank group,with 20 rats in each group.Rats in the model group and experiment group were injected with rotenone in the right substantia nigra pars compacta(SNc)region and right ventral tegmental area(VTA)of midbrain to establish PD model with unilateral complete damage.Then PD rats in the experiment group and rats in the virus group were injected with adeno-associated virus.The rats in the blank group were not given any intervention.The SNc region of rats in the four groups were implanted with photoelectric pole for giving light stimulation and then the electroencephalogram(EEG)signals were collected before,during and after light stimulation,and the changes of EEG signal were analyzed by LZ complexity.Results There was a significant difference in the LZ complexity between PD rats and normal rats.The optogenetics has a significant effect on the EEG signal of rats in the experiment group and the virus group,but it had no effect on the EEG signal of rats in the model group and the blank group.Theθwave of EEG of rats in the experiment group was more similar to that in the blank group when the rats were stimulated by light.Conclusion Optogenetics technique has therapeutic effect on PD rats.
