长链非编码RNA LINC00963在喉鳞状细胞癌中的表达及其对Hep-2细胞增殖及凋亡的影响

Expression of LINC00963 in laryngeal squamous cell carcinoma and its effects on proliferation and apoptosis of Hep-2 cells in vitro and in vivo

目的检测基因间长链非编码RNA 00963(LINC00963)在喉鳞状细胞癌组织及喉癌细胞Hep-2中的表达及其对Hep-2细胞增殖及凋亡的影响。方法聚合酶链式反应和免疫组化检测喉鳞状细胞癌组织及Hep-2细胞中LINC00963的表达水平;CCK-8及流式细胞术体外水平检测沉默LINC00963表达对细胞增殖及细胞凋亡的影响;荷瘤裸鼠实验检测LINC00963对肿瘤生长的影响;Ki-67及TUNEL染色体内水平检测LINC00963对Hep-2细胞增殖及凋亡的影响。结果41例喉鳞状细胞癌组织中LINC00963的表达水平高于癌旁组织。LINC00963在Hep-2细胞中的表达也明显高于永生化鼻咽上皮NP-69细胞。沉默Hep-2细胞中LINC00963的表达后,细胞增殖速度下降,凋亡细胞比例增高(P<0.05)。此外,与LINC00963 NC组相比,LINC00963 shRNA组细胞荷瘤裸鼠肿瘤生长明显受到抑制、Ki-67阳性细胞比例降低、TUNEL阳性细胞比例增高(P<0.05)。结论LINC00963在喉鳞状细胞癌组织及细胞中异常高表达,并可在体内及体外水平促进Hep-2细胞增殖、抑制细胞凋亡。

Objective To investigate the expression of long intergenic non-coding RNA 00963(LINC00963)in tissues and cells of laryngeal squamous cell carcinoma(LSCC)and explore the effects of LINC00963 on cell proliferation and apoptosis of Hep-2 cells.Methods QPCR and immunohistochemistry were performed to detect the expression of LINC00963 in tissues and cells of laryngeal squamous carcinoma.CCK-8 and flow cytometry assays were performed to detect the effects of LINC00963 on cell proliferation and apoptosis of Hep-2 cells in vitro.Xenograft assay of Hep-2 cells was performed to detect the effects of LINC00963 on tumor growth.Ki-67 and TUNEL assays were performed to detect the effects of LINC00963 on cell proliferation and cell apoptosis in vivo.Results The level of LINC00963 in 41 LSCC tissues was upregulated compared with that of adjacent tissues.Compared to immortalized nasopharyngeal epithelial cell line,NP-69 cells,LINC00963 was also upregulated in LSCC cell line,Hep-2.After silencing the expression of LINC00963 in Hep-2 cells,cell proliferation was inhibited and cell apoptosis was promoted in vitro(P<0.05).Furthermore,compared to Hep-2 cells transfected with LINC00963 NC,tumor growth was suppressed,Ki-67 positive cells were down-regulated,and TUNEL positive cells were up-regulated in vivo(P<0.05).Conclusion LINC00963 is up-regulated in tissues and cells of LSCC.LINC00963 can promote cell proliferation and inhibit cell apoptosis of Hep-2 cells in vitro and in vivo.
