摘要
目的采用正交试验设计优化微波结合内生真菌发酵液浸泡辅助水蒸气蒸馏法提取马来沉香挥发油的提取工艺。方法对真菌发酵液浸泡时间、料液比及微波时间3个因素进行考察,将不同工艺所得挥发油的乙醚萃取液进行GC-MS分析,以总峰面积和倍半萜、芳香族类化合物的总峰面积作为综合指标,优化最佳工艺条件。结果真菌发酵液浸泡时间对沉香挥发油提取工艺具有显著影响,最佳工艺参数为发酵液浸泡4 d,料液比1∶8,微波时间对提取没有影响。结论真菌发酵液浸泡辅助水蒸气蒸馏法可用于沉香挥发油的提取。
Objective Objective Orthogonal design was used to optimize the extraction process of volatile oil from Aquilaria malaccensis by microwave combined with fungal fermentation broth immersion and auxiliary steam distillation.Methods The three factors of immersion time(A),solid-liquid ratio(B)and microwave time(C)were investigated.The ether extract obtained by different processes were analyzed by GC-MS.The total peak area and the total peak area of sesquiterpene and aromatic compounds were used as comprehensive indicators to analyze the optimum process conditions.Results Immersion time of agarwood in fermentation broth had a significant effect on the extraction process.The best process parameters were immersion time of 4 days and the ratio of material to liquid of 1:8.Microwave time had no effect on the extraction process.Conclusion Fungal fermentation borth immersion and auxiliary steam distillation can be used for the extraction of agarwood volatile oil.
