摘要
目的采用GC-MS技术对梅归参颗粒挥发油进行成分分析,确定方剂中挥发油的化学成分,研究其体外抑菌作用,为梅归参颗粒的临床应用提供实验数据。方法用水蒸气蒸馏法提取梅归参颗粒挥发油,采用GC-MS技术对其化学成分进行分析鉴定,并进行体外抑菌作用研究。结果GC-MS共分离出可识别峰138个,鉴定出相对质量分数≥0.1%的化学成分有25种,其中相对质量分数>1.0%的化学成分有8种。体外抑菌实验表明,梅归参颗粒挥发油对铜绿假单胞菌、大肠埃希菌、金黄色葡萄球菌、变形杆菌、乙型溶血性链球菌、伤寒杆菌均有显著的抑制作用,MIC分别为1.86、3.73、14.91、3.73、1.86、7.45 mg/mL,MBC分别为7.45、14.90、7.45、3.73、3.73、14.90 mg/mL。结论梅归参颗粒挥发油具有显著的抗菌作用,研究结果为梅归参的临床应用提供了依据,建立的方法学为梅归参颗粒质量标准提高提供了参考。
Objective The chemical composition of the essential oils in Meiguishen granules was determined by GC-MS.Antibacterial activity in vitro was studied in order to provide experimental data for the clinical application of meiguishen granules.Methods The essential oils of meiguishen granules was extracted by steam distillation,and the chemical composition of the essential oils in Meiguishen granules was identified by GC-MS.In addition,the antibacterial effect in vitro was studied.Results A total of 138 peaks were identified by GC-MS.The relative content of 25 compounds was higher than 0.1%.Among them,there were 8 components with relative contents larger than 1.0%.in vitro,The essential oils in Meiguishen granules showed strong inhibition against all tested bacteria(Pseudomonas aeruginosa,Escherichia coli,Staphylococcus aureus,Proteus,Streptococcus hemolyticus type B,Salmonella typhi),with MIC(1.86,3.73,14.91,3.73,1.86,7.45 mg/mL)and MBC(7.45,14.90,7.45,3.73,3.73,14.90 mg/mL)values.Conclusion This study indicates that the essential oils of Meiguishen granules possess strong antibacterial properties against the six strains pathogenic bacteria,which provides experimental support for the clinical application of Meiguishen granules.Furthermore,the established methodology provides a reference for improving the quality standard of Meiguishen granules.
