姜黄素调节β-catenin/TCF4通路对胃癌细胞体内外生长和转移的影响 被引量:6

Effect of curcumin on growth and metastasis of gastric cancer cells in vitro and in vivo by regulating β-catenin/TCF4 pathway

摘要 目的:探讨姜黄素(CM)对胃癌MGC-803细胞增殖、凋亡、侵袭和上皮间质转化的调节作用及其机制。方法:将细胞分为Control组、CM(10μmol/L)组、CM(20μmol/L)组和CM(40μmol/L)组,检测胃癌MGC-803细胞增殖、凋亡、侵袭、迁移和上皮间质转化(EMT)能力和相关蛋白表达情况;建立胃癌异种移植瘤模型,检测小鼠存活率、肿瘤体积、肿瘤细胞凋亡率和Ki67表达量。结果:与对照组相比,CM剂量依赖性抑制胃癌MGC-803细胞增殖和侵袭转移,促进细胞凋亡,且增加移植瘤小鼠存活率和细胞凋亡率、降低肿瘤体积和组织中Ki67表达水平。结论:CM能抑制胃癌MGC-803细胞体内外增殖、侵袭和间质转化能力,诱导凋亡,其机制与抑制β-catenin/TCF4通路活化有关。 Objective:To investigate the regulatory effect of curcumin(CM) on proliferation,apoptosis,invasion and epithelial-mesenchymal transition of gastric cancer MGC-803 cells and its mechanism.Methods:Cells were divided into control group,CM(10 μmol/L) group,CM(20 μmol/L)group and CM(40 μmol/L)group.Proliferation,apoptosis,invasion,migration and epithelial-mesenchymal transition(EMT) ability and related protein expression of MGC-803 cells were detected.The xenograft model of gastric cancer was established,and the survival rate,tumor volume,apoptotic rate of tumor cells and expression of Ki67 in mice were detected.Results:Compared with the control group,CM dose-dependent inhibited the proliferation,invasion and metastasis of gastric cancer MGC-803 cells,promoted cell apoptosis,increased the survival rate and apoptosis rate of transplanted tumor mice,and decreased the volume of tumor and Ki67 expression level in tissues.Conclusion:CM inhibits proliferation,invasion,mesenchymal transformation and induce apoptosis of gastric cancer MGC-803 cells in vitro and in vivo.Its mechanism is related to inhibiting activation of β-catenin/TCF4 pathway.
