化瘀解毒方对胃癌上皮间质转化作用的研究

目的:探讨化瘀解毒方(HYJD)对TGF-β1诱导的人胃癌SGC-7901细胞上皮间质转化(EMT)的影响,为化瘀解毒方的临床应用及进一步成药开发提供理论和实验基础。方法:运用细胞因子TGF-β1诱导人胃癌SGC-7901细胞进行EMT造模,同时设置阴性对照组、模型组和不同浓度解毒化瘀方处理实验组。MTT法检测解毒化瘀方对胃癌细胞生长抑制作用;Western blot法检测细胞EMT生物标志分子(Bio-marker)E-钙黏蛋白(E-cadherin)、N型钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)的表达水平;Transwell小室法用来检测胃癌细胞体外的侵袭和转移能力。结果:TGF-β1能够成功诱导SGC-7901细胞发生EMT,使细胞的形态发生改变。与模型组相比:HYJD能显著抑制胃癌细胞的增殖,呈明显的剂量依赖性。HYJD能下调人胃癌SGC-7901细胞的EMT标志蛋白N-钙黏蛋白、波形蛋白的表达水平,同时上调E-钙黏蛋白的表达水平,显著抑制人胃癌SGC-7901细胞的EMT进程,呈明显的剂量依赖性;HYJD能降低人胃癌SGC-7901细胞的穿膜细胞数。结论:HYJD能明显抑制人胃癌SGC-7901细胞的增殖,同时可以抑制胃癌细胞的EMT进程并逆转SGC-7901细胞的EMT进程,从而抑制胃癌细胞的侵袭转移。

Objective:To study the effect of Huayu Jiedu Recipe(HYJD) on suppressing the epithelial-mesenchymal transition(EMT) of human gastric cancer cells.Methods:The epithelial-mesenchymal transition of SGC-7901 cells were induced by TGF-β1.We set model group,negative control group and different concentrations of HYJD groups.MTT assay was used to detect the inhibitory effect of HYJD on the growth of gastric cancer cells.Western blot was used to detect the expression levels of EMT biomarker(Bio-marker),E-cadherin,N-cadherin and Vimentin.Invasion assay and migration assay were used to observe the effect of HYJD on the metastatic ability of gastric cancer cells in vitro.Results:TGF-beta 1 can successfully induce EMT in SGC-7901 cells,and change the cell morphology.Compared with the EMT model group,SGC-7901 cells treated with HYJD at different concentrations exhibited a dose-dependent growth inhibition.In addition,the expression levels of EMT Bio-marker N-cadherin and Vimentin were prominently suppressed by HYJD in a concentration-dependent manner,while the expression level of E-cadherin were raised.HYJD obviously reduced the invasion potential of SGC-7901 cells.Conclusion:HYJD can significantly inhibit the proliferation of human gastric cancer SGC-7901 cells.HYJD can inhibit the EMT process of gastric cancer cells and reverse the EMT process of SGC-7901 cells,thus inhibiting the invasion and metastasis of gastric cancer cells.
