咳喘宁对RSV诱发哮喘豚鼠肺TLR4及NF-κB mRNA的影响 被引量:1

摘要 目的:研究咳喘宁对RSV诱发哮喘豚鼠肺组织TLR4及NF-κB mRNA表达的影响,探讨其在哮喘治疗中的作用及机制。方法:60只清洁级豚鼠中随机抽取10只作为空白组,余下50只均用呼吸道合胞病毒(RSV)滴鼻加卵清蛋白(OVA)雾化吸入进行造模。造模成功后按随机数字表法平均分为模型组,咳喘宁高、中、低剂量组,西药对照组各每组10只。进行相应的处理后留取肺组织标本HE染色后观察气道炎症,实时荧光定量PCR法检测豚鼠肺组织TLR4及NF-κB mRNA的表达。结果:与空白对照组比较,模型组豚鼠支气管管壁明显增厚,结构明显紊乱,有炎症细胞浸润;与模型组比较,药物治疗组豚鼠肺组织的病理变化均有所改善。与空白对照组比较,模型组豚鼠肺组织中TLR4及NF-κB mRNA的表达水平显著增高,差异有统计学意义(P<0.05)。与模型组比较,药物治疗组豚鼠肺组织中的TLR4及NF-κB mRNA表达水平均显著降低,差异有统计学意义(P<0.05)。结论:咳喘宁可以通过下调TLR4、NF-κB的表达水平,减缓RSV诱发哮喘豚鼠肺组织气道重塑,减轻炎症反应。 Objective:To study the effect of Kechuanning on the expression of TLR4 and NF-κB mRNA in lung tissue of guinea pigs with asthma induced by RSV,and to explore its role and mechanism in the treatment of asthma.Method:10 clean guinea pigs were randomly selected as the blank group,and the remaining 50 were modeled with respiratory syncytial virus(RSV)nasal drops and ovalbumin(OVA)aerosol inhalation.After successful modeling,according to the random number table method,they were evenly divided into model group,high,medium,and low dose of Kechuanning groups and western medicine control group,with 10 guinea pigs in each group.After the corresponding treatment,lung tissue specimens were left for HE staining and airway inflammation was observed.Realtime fluorescent quantitative PCR was used to detect the expression of TLR4 and NF-κB mRNA in guinea pig lung tissue.Result:Compared with the blank control group,the bronchial wall of the guinea pigs in the model group was significantly thickened,the structure was significantly disordered,and inflammatory cells were infiltrated.Compared with the model group,the pathological changes in the lung tissue of the guinea pigs in the drug treatment group were improved.Compared with the blank control group,the expression levels of TLR4 and NF-κB mRNA in the lung tissue of guinea pigs in the model group were significantly increased,and the difference was statistically significant(P<0.05).Compared with the model group,the expression level of TLR4 and NF-κB mRNA in lung tissue of guinea pigs in the drug treatment group were significantly reduced,and the difference was statistically significant(P<0.05).Conclusion:Kechuanning can slow down RSV-induced airway remodeling in lung tissue of guinea pigs by reducing the expression levels of TLR4 and NF-κB,and reduce inflammation.
