抑癌基因Reprimo启动子区甲基化在胰腺癌中的表达

摘要 目的:检测胰腺癌中抑癌基因Reprimo启动子区甲基化水平及蛋白的表达,探讨其与胰腺癌发生发展的关系。方法:选取48例胰腺癌组织及18例癌旁组织为研究对象。免疫组化及半定量RT-PCR技术检测Reprimo在两种组织中的表达;甲基化特异聚合酶链反应(MSP法)检测其启动子区甲基化水平;分析Reprimo的表达与其启动子区甲基化水平的关系。结果:与癌旁组织相比较,Reprimo mRNA在胰腺癌组织中的表达明显降低,差异有统计学意义(P<0.05);Reprimo蛋白在胰腺癌组织及癌旁组织中的表达率分别为16.67%(8/48)和83.33%(15/18),差异有统计学意义(P<0.05);胰腺癌组织中Reprimo启动子区甲基化率为79.17%(38/48),癌旁组织中未检测出Reprimo启动子区甲基化,差异有统计学意义(P<0.001);Pearson列联系数分析显示Reprimo蛋白的表达与其启动子区甲基化率呈负相关(r=-0.298,P<0.05)。结论:Reprimo基因启动子区异常甲基化使Reprimo表达水平降低,可能参与胰腺癌的发生发展。 Objective:To study the promoter methylation of cancer suppressor gene Reprimo and its clinical value in pancreatic cancer.Methods:Forty eight cases of pancreatic cancers and 18 cases of normal pancreatic tissues were selected to perform this study.The protein and mRNA expression of Reprimo were detected by immunohistochemistry and semi quantitative RT-PCR technique.The methylation level of Reprimo gene was detected by methylation specific PCR(MSP)method.The relationship between the expression of Reprimo and promoter methylation was analyzed.Results:Compared with the normal tissue,the mRNA expression of Reprimo was significantly lower in pancreatic cancer(P<0.05).The expression rate of Reprimo in pancreatic cancer tissue and normal tissue were 16.67%(8/48)and 83.33%(15/18),with a significant difference(P<0.05).The promoter region methylation rate was 79.17%(38/48)in pancreatic cancer tissue,but undetected in normal tissue.The expression Reprimo was negatively correlated to promoter methylation(r=-0.298,P<0.05).Conclusion:The Reprimo promoter abnormal methylation can reduce the expression of Reprimo,which may participate in the occurrence and development of pancreatic cancer.
