摘要
无标记细胞分析法(RTCA)与CCK-8法,可用于奶牛乳腺上皮细胞(MAC-T)活性监测。本研究选择RTCA和CCK-8两种方法,在0、5、10、20ng/μL4种不同浓度LPS条件下,研究LPS对MAC-T增殖的影响。结果表明,RTCA和CCK-8法均在不同浓度LPS条件下使MAC-T活力下降,均在浓度20ng/μL、时间6h时,下降最为明显。但相比之下,CCK-8法在定点测定时有局限性,而RTCA方法可以不间断、无标记和实时监测细胞贴壁、迁移和增殖的动态等生物反应过程,观察效果更加直观、准确,可较好地弥补CCK-8法的缺点,是较理想的研究奶牛乳腺上皮细胞增殖的方法。
To compare the activity of bovine mammary epithelial cells(MAC-T)by real time cell analysis(RTCA)and CCK-8 method to determine the best experimental method for studying the effect of LPS on cell proliferation.CCK-8 method and real time cell analysis were used to determine the effect of LPS on the proliferation of MAC-T cells.The results of CCK-8 and real time cell analysis(RTCA)showed that different concentrations of LPS decreased the viability of MAC-T cells,and the most significant decrease was observed at the concentration of 20 ng/uL and at the time of 6 h.But real time cell analysis(RTCA)can monitor the dynamic biological reaction process of cell adherence,migration and proliferation without interruption,unlabeled and real time,and the observation effect is more intuitive and accurate.Real time cell analysis(RTCA)can make up for the shortcomings of CCK-8 method and become one useful method to investigate cell proliferation.
