摘要
目的观察低氧/冷暴露对肥胖大鼠白色脂肪组织中UCP-1表达的影响,并讨论该复合环境对白色脂肪棕色化是否有增强效应。方法将20只肥胖大鼠随机分为正常对照组(N,n=5)、常温/低氧组(NT/H,n=5)、常氧/冷暴露(NH/C,n=5)及低氧/冷暴露组(H/C,n=5);常温/低氧组及低氧/冷暴露组在模拟海拔5500m的低压氧仓造模;四组大鼠均正常饲养四周,并对大鼠体重进行称量,应用Q-PCR技术检测大鼠肾周白色脂肪组织中UCP-1的表达。结果(1)体重变化统计结果:①正常对照组大鼠体重实验前后无明显变化;②与正常对照组相比较,低氧/冷暴露组大鼠体重显著降低,差别有统计学意义(P<0.05);③与低氧/冷暴露组相比较,低氧/常温组和常氧/冷暴露组大鼠的体重出现下降趋势,差异有统计学意义(P<0.05);(2)UCP-1表达量统计结果:①正常对照组大鼠白色脂肪组织中UCP-1水平的表达实验前后无明显变化(P>0.05);②低氧/冷暴露组大鼠白色脂肪组织中UCP-1表达较正常对照组显著增加(P<0.05);③与低氧/冷暴露组相比,低氧/常温组和常氧/冷暴露组大鼠白色脂肪组织中UCP-1表达有所增加,差异有统计学意义(P<0.05)。结论经过低氧/冷暴露干预后,随着肥胖大鼠白色脂肪组织UCP-1表达量的增高,体重不断减轻,说明低氧冷暴露可能是通过上调UCP-1的表达量进而影响体重。
Objective To observe the effects of hypoxia/cold exposure on the expression of ucp-1 in white adipose tissue of obese rats,and to discuss whether there has enhancement effects of the compound environment on browning of white fat.Methods 20 obese rats were randomly allocated to normal control group(N,n=5),normal temperature/hypoxia group(NT/H,n=5),normoxia/cold exposure group(NH/C:n=5)and hypoxia/cold exposure group(H/C:n=5),The normal temperature/hypoxia group and hypoxia/cold exposure group were modeled in low-pressure oxygen warehouse with simulated altitude of 5500m;fed for 4 weeks in different environment,and weighed the rats,detected the expression of ucp-1 gene in perirenal white adipose tissue of rats by Q-PCR.Results 1)Statistical results of weight change:①There was no significant changes of the weight in normal control group before and after the experiment;②Compared with the normal control group,the weight of hypoxia/cold-exposed decreased significantly,the difference was statistically significant(P<0.05);③Compared with the hypoxia/cold exposed group,the weight of the hypoxia/normal temperature group and the normoxia/cold exposed group showed a downward trend,the difference was statistically significant(P<0.05);2)Statistical results of UCP-1 expression:①the expression of ucp-1 in white adipose tissue of rats in the normal control group showed no significant change before and after the experiment(P>0.05);②The expression of Ucp-1 in white adipose tissue of hypoxia/cold group was significantly increased compared with that of normal control group(P<0.05);③Compared with hypoxia/cold exposure group,The expression of ucp-1 in white adipose tissue of hypoxia/normal temperature group and normoxis/cold exposure group was increased,with a significant difference(P<0.05).Conclusion After hypoxia/cold exposure intervention,with the increase of ucp-1 expression in white adipose tissue of obese rats,the body weight was continuously reduced,indicating that hypoxia and cold exposure may upregulate the body weight by affecting the expression of ucp-1.
