六味地黄汤对去卵巢绝经后骨质疏松症模型大鼠的早期干预作用 被引量:16

摘要 目的:观察六味地黄汤对去卵巢绝经后骨质疏松症模型大鼠补肾的早期干预作用及其机理。方法:将30只大鼠采用去卵巢的方法构建绝经后骨质疏松症动物模型,随机分为模型组、六味地黄汤组、阿仑膦酸钠片组,另设假手术组,共4组。造模后第5天开始,假手术组、模型组灌服生理盐水,其余各组按药物相应剂量灌胃给药,且每周各组称质量1次。术后90 d,检测模型大鼠骨密度(bone mineral density,BMD)值及血清雌二醇(estradiol,E2)、钙(calcium,Ca)、磷(phosphorus,P)水平,镜下观察骨组织结构。结果:与模型组比较,六味地黄汤组可以升高模型大鼠BMD值及血清E2、Ca、P水平,减少骨小梁的断裂及骨髓腔中脂肪细胞的含量,并抑制模型大鼠体质量的持续增长而致肥胖。结论:六味地黄汤对绝经后骨质疏松症前骨亚健康状态补肾的早期干预作用可以延缓绝经后骨质疏松症的发生发展。其主要作用机理与六味地黄汤升高雌激素水平,减缓骨中Ca、P的丢失,进而减少骨小梁的断裂及骨髓中脂肪细胞的含量,或抑制体质量增长而致的肥胖,延缓BMD值下降有关。 Objective:To observe the early intervention effect and mechanism of Liuwei Dihuang Decoction on tonifying kidney in ovariectomized postmenopausal osteoporosis model rats.Methods:Thirty rats were used to construct an animal model of postmenopausal osteoporosis by ovarian degeneration.They were randomly divided into model group,Liuwei Dihuang Pill group,alendronate sodium group,and sham operation group,a total of 4 group.On the 5 th day after modeling,the sham operation group and the model group were given normal saline,and the other groups were intragastrically administered according to the corresponding dose of the drug,and each group was weighed once a week.At 90 o′clock,the Bone mineral density(BMD)value and serum estradiol(E2),calcium(calcium,Ca),phosphorus(phosphorus,P)and level and organizational structure were observed and measured.Results:Compared with that of the model group,Liuwei Dihuang Decoction group can increase the BMD value and serum E2,Ca,P levels in model rats,reduce the fracture of trabecular bone and the content of fat cells in the bone marrow cavity,and inhibit the model rats.Obesity caused by the continued increase in body mass.Conclusion:Liuwei Dihuang Decoction can delay the development of postmenopausal osteoporosis in the early intervention of tonifying kidney before the postmenopausal osteoporosis.Its main mechanism of action is related to Liuwei Dihuang Decoction increasing the estrogen levels to slow the loss of Ca and P in bone minerals,thereby reducing the fracture of trabecular bone and the content of fat cells in bone marrow,or inhibiting the increase of body mass.,delaying the decline in BMD values.
